| Objective:Human lung adenocarcinoma A549cells were disposed by curcumin, observedthe change of the cell morphology, detected the cell cycle, cell proliferation, apoptosisand activity of ubiquitin-protease in A549cells, to investigate the effect of curcuminon biological behavior of A549cells and related molecular mechanism, which wouldprovide theoretical basis and new approach for the clinical application of lung cancer.Methods:1. Established the lung adenocarcinoma A549passage cell model and disposedwith curcumin by concentration of0umol/L,10umol/L,20umol/L,40umol/L and80umol/L for24h,48h,72h, observed the changes of cell morphology under invertedmicroscope, detected the proliferation inhibition of A549cell by MTT assay andcalculated the IC50of curcumin in different period.2. Disposed A549cells with curcumin according to its IC50, detected the effectof curcumin on the cell cycle in lung cancer by FCM.3. Disposed the lung adenocarcinoma A549cells with0umol/Lã€10umol/Lã€20umol/Lã€40umol/L concentrations of curcumin for12h,24h and48h, detected theeffect of curcumin on apoptosis in A549cells by FCM.4. Detected the expression of protein Skp2, cul1(both are the structure of SCFubiquitin ligase) and P27(a protein regulate cell cycle) in A549cells after diaposed ofdifferent concentrations of curcumin at different time period by Western blot.Results:1. Curcumin could inhibit the proliferation of lung adenocarcinoma A549cellsin a time and the concentration dependent manner. The48h and72h IC50ofcurcumin were27.98umol/L,20.98umol/L respectively.2. Curcumin could induce cell cycle arrest at G1phase in a time dependentmanner.3. Curcumin could promote the apoptosis of lung adenocarcinoma A549cells ina time and the concentration dependent manner. 4. Curcumin could increase the expression of P27but decrease the expression ofthe structure protein of SCF ubiquitin ligase Skp2and cul1in A549cells.Conclusion:Curcumin may inhibit the proliferation of lung cancer A549cells, arrest its cellcycle at G1phase and induce its apoptosis by down-regulating the expression of SCFubiquitin ligase, which can provide a new approach for the prevention and treatmentof lung cancer. |
