Methods:96female BALB/c mice (age of6~8weeks,18-22g), through slit lampmicroscope, eye anterior segment and fundus ophthalmoscope examination and therewas no apparent unusual. The mice were randomly divided into6groups, each groupof16, except the blank control group (BC), others adopt to be dry eye induced byhypertonic saline for14days, through tear secretion(Schirmer I), cornea fluorescentstaining to decide which would to be pushed out of the control group, aftersuccessfully inducted, mice were randomly divided into the hypertonic saline eyedrops group (HO), high permeability joint high dose of Chinese traditional medicinegroup (HO+HD group), high permeability joint middle dose of Chinese medicinegroup (HO+MD group), high permeability joint low dose of Chinese traditionalmedicine group (HO+LD group), high permeability joint hyaluronate sodium eyedrops group (HO+SH group). MD+HD group、HO+MD、HO+LD group startlavaging Yi Qi Run Mu Decoction for14days,1/d. HO+SH Group with hyaluronatesodium eye drops for14days,3/d.8mice were killed of each group when havegavaged7days and14days, then observe the change through paraffin pathologicalsection, HE staining, PAS staining,immunohistochemistry, and reversetranscription-polymerase chain reaction.Results:1.With14day’s hypertonic saline drops, except the blank group, tearsecretion of other mice decrease significantly, and it also appears the increased corneafluorescent staining,P<0.05, the difference was statistically significant;2.It showsthat the tear secretion of the groups with traditional Chinese medicine significantlyincreased and it also appears reduced dyeing score after treatment, P<0.05, comparedwith HO group the difference is statistically significant; Pathological HE staining: thecorneal epithelial thickness of HO group has reduced, the corneal epithelial layerdecreased, the arrangement of cells disordered and very free bubble changement,however other groups have no obvious change. The PAS staining: show that thenumber of fornix conjunctival goblet cells of Chinese traditional medicine group is obviously increased compared with HO group; Immunohistochemical confirm: With7and14days’ treatment the expression of IL-1β in the cornea is lower with Chinesetraditional medicine group than HO group (P<0.05), as while reverse transcriptionfollowed by PCR brough out TCM group has lower expression of IL-1β than HOgroup,the difference is statistically significant.Conclusion:1.The mice induced by hypertonic saline showed the sign and symptomof dry eye;2.Yi Qi Run Mu decoction can effectively stimulate the tear secretion,maintain the tear film stability, improve dry eye symptom which probably due toinhibiting the expression of inflammation factor in ocular surface.
