Targets:1. Explore the effect of Curcumin Preconditioning on Behaviour science of Acute Spinal Cord Injury in mice.2. Explore the effect of Curcumin Preconditioning on Pathology of Acute Spinal Cord Injury in mice.3. Explore the effect of Curcumin Preconditioning on Cytokines of Acute Spinal Cord Injury in mice.4. Provide theory basis for clinical treatment on Acute Spinal Cord Injury.Methods:1. Establish experimental animal models following Improved Allen’s Model.2. Divide24healthy and clean class mice into Normal team, Model team, MP(methylprednisolone) team and Curcumin team, and inject relative medicines accordingly before establishment of Spinal Cord Injury model. Perform behavior evaluation on experiment mice with Nerve function BMS grading and RIVLIN inclined plate failure rate after24h,48h and72h of experiment.3. Divide96healthy and clean class mice into Normal team, Model team, MP(methylprednisolone) team and Curcumin team, and inject relative medicines accordingly before establishment of Spinal Cord Injury model. After8h,24h,48h, and72h of experiment, make HE staining section and observe through light microscope. Examine Nrf2antibody expression with Western blot method, and expression of TNF-α and IL-6with Elisa method.Results:1. Mice behavior science: neurologic function of Curcumin team achieved significant improvement compared to Model team (P,<0.05), but relatively lower compared to MP(methylprednisolone) team.2. HE staning tissue Pathology: the injured spinal cord tissue of Curcumin team revealed visible hemorrhage, Neuronal cell size turned down, the nucleolus become unclear or disappeared, and the infiltration of inflammatory cells. The degree of injury was less serious compared to Model team.3. Western blot Test found only limited expression of Nrf2in Model team, the expression in Curcumin team is significantly stronger compared to Model team in each time spot, though still lower compared to MP(methylprednisolone) team.4. Elisa Test found TNF-α expression in Curcumin team in different time spot is lower than Model Team (P<0.05) in8h、24h and72h, but higher than MP(methylprednisolone) team. IL-6expression in Curcumin team only revealed weaker in8h compared to Model team (P<0.05), but no notable difference compared to Model team (P<0.05) after8h..Conclusion:Curcumin Preconditioning before operation can strengthen behavior ability of mice with Acute Spinal Cord Injury, and improve the Pathology change of Spinal Cord Injury in some extent. Infiltration of inflammatory cells, Nrf2expression raise are also found in experiment, stating the improvement on neurologic function recovery. The preliminary study shows that Curcumin Preconditioning brings preventive effect in a certain level.
