Objective1.Gal-1expression of liver resection specimens was analyzed clinicallyand correlation with clinically relevant datas was statisticed;2.Low Gal-1expressioninterference plasmid whose interference effect was verified was constructed to be thefoundation of construction of interference cell lines;3.After success in building lowexpression or over-expression Gal-1of eukaryotic plasmids,high expression Gal-1ofhepatoma cell line was selected for the construction of low expression Gal-1cell lineand initial low expression Gal-1of hepatoma cell lines was selected for theconstruction of high expression Gal-1cell line,it laied the foundation for the lateranalysis of the biological effects of Gal-1in liver cancer cell lines;4.The effects ofGal-1on constructed hepatoma cell lines was analyzed,such as the effects on HCCmigration, invasion and so on, and the mechanism was explored briefly.Method1. The protein of62cases of clinical specimens of primary liver cancerwas extracted, western blot technology was used to analyze the expression of Gal-1inthese clinical samples, the gray value of Gal-1was analyzed by gray value analysissoftware, then the gray value was divide into high and low groups, relevant clinicaldata collected, and then the significance of correlation between Gal-1and clinical datawas analyzed by the related statistical methods;2. Related Gal-1interferencesequence was searched through literature, and then corresponding interferencesequence was synthesised, the DNA recombinant techniques was used to constructGal-1interference or overexpression of eukaryotic plasmids;3. The constructedplasmids was transfect into hepatoma cell lines by transfection reagent, the stabletransfection of a single clone was gotten through drug screening,then was amplifiedto stably transfected cell lines;4.on the basis of the success construction of hepatomacell lines, the impact of Gal-1was analyzed on the migration, invasion of hepatomacell line, and the possible mechanism probed..Results1.Through analysis of the relationship between Gal-1expression in clinicalspecimens of62cases of primary liver cancer and quatifying clinical datas, the poordifferentiation,large tumor volume、 high metastasis was found in high Gal-1 expression liver cancer than in low Gal-1expression liver cancer;2.By plasmidsequencing and transient trasfection, eukaryotic interference plasmid sequences wasfound to be collect, and the interference effects is ok;3.Through drug screening, theconcentration of drug using for screening cell lines is clear, and clear interferenceeffect of cell lines was constructed successfully;4.Through the analysis of the Gal-1interference cell line,we found that the migration and invasion abilities of the cell linewere weakened, and Gal-1recombinant protein (r-Gal) and Gal-1overexpression cellline were both able to promote their abilities of migration and invasion, also foundthat Gal-1promoted hepatocellular carcinoma cell lines undergo epithelial-mesenchymal transition.Conclusion1.Gal-1is associated with growth and metastasis of liver cancer;2.Gal-1interference plasmid was constructed correctly;3.Hepatoma cell lines whichexpresses low and high Gal-1were constructed successfully;4.Gal-1promote livercancer cell migrate, invade, and induce EMT changes in hepatoma cells.
