| ObjectivesThe objective of the study was to observe the effects of different dietary ironcontents on the hematological iron status, the hippocampal non-heme-iron (NHI)contents and other metal ions (sodium, magnesium, potassium, manganese, copperand zinc) contents in male rats, and observe the effects of long-term exercise on thehematological iron status, the contents of NHI and other metal ions (sodium,magnesium, potassium, manganese, copper and zinc) in the hippocampus withdifferent dietary iron contents.MethodsOne hundred and ten weaning male rats were assigned into three groups: adietary iron deficiency group (12mg/kg), a dietary iron sufficiency group (45mg/kg)and a high dietary iron content group (1000mg/kg). The rats were fed withcorresponding iron content diets for one month. Fifteen rats of each group were fastedfor24hours, and then blood samples were taken when rats were anesthetized withpentobarbital. The blood samples were collected to determine the hematologicalindices of iron status (Hb, HCT, MCV, MCH, RDW, SI, TIBC, TS). The brain wasremoved immediately,and the hippocampus was isolated to determine the contents ofNHI, sodium, magnesium, potassium, manganese, calcium, copper or zinc. And then, the rats of each group were further divided into an exercise group and a sedentarygroup: exercise group with dietary iron deficiency, sedentary group with dietary irondeficiency, exercise group with dietary iron sufficiency, sedentary group with dietaryiron sufficiency, exercise group with high dietary iron content, sedentary group withhigh dietary iron content. The rats in three exercise groups were exposed to swimmingexercise for three months (5d/week, one exercise session/d), and the rats in thecorresponding sedentary groups were subjected to the same treatments exceptswimming. Animals were fasted for24hours after the last exercise regimen and thehematological indices of iron status (Hb, HCT, MCV, MCH, RDW, SI, TIBC, TS), thecontents of NHI, sodium, magnesium, potassium, manganese, copper or zinc in thehippocampus were determined.Results1. The impact of dietary iron contents on the hematological indices of ironstatus, hippocampal iron status and other metal ions contents in male ratsDietary iron contents could affect Hb, HCT, MCV, SI, TIBC and TS (P<0.05).Compared to CN group, Hb, HCT, MCV, MCH, SI and TS were significantlydecreased (P<0.01), RDW and TIBC were significantly increased (P<0.01) in IDgroup. Compared to CN group, Hb, HCT and MCV were increased (P<0.05), SI andTS were significantly increased (P<0.01), TIBC significantly decreased (P<0.01),MCH and RDW had no difference (P>0.05) in SU group.Dietary iron contents could not affect the contents of NHI, sodium, magnesium,potassium, manganese, and zinc in the hippocampus (P>0.05). In ID group thecontents of copper in the hippocampus was increased (P<0.05).2. The impact of long-term exercise on the hematological indices of iron status,hippocampal iron status and other metal ions contents in male rats with differentdietary iron contentsCompared to sedentary group with dietary iron deficiency, TIBC and zinccontents in the hippocampus were increased (P<0.05), TS and NHI contents in thehippocampus were decreased (P<0.05), Hb, MCV, MCH, HCT, RDW, SI and othermetal ions (sodium, magnesium, potassium, manganese, and copper)contents had no difference (P>0.05) in exercise group with dietary iron deficiency.Compared to sedentary group with dietary iron sufficiency, RDW and coppercontents in the hippocampus were significantly increased (P<0.01), Hb, HCT, MCV,MCH, SI, TIBC, TS, the contents of NHI, sodium, magnesium, potassium, manganese,and zinc in the hippocampus had no difference (P>0.05) in exercise group withdietary iron sufficiency.Compared to sedentary group with high dietary iron content, RDW wassignificantly increased (P<0.01), Hb, HCT, MCV, MCH, SI, TIBC, TS, the contentsof NHI, sodium, magnesium, potassium, manganese, copper and zinc in thehippocampus had no difference (P>0.05).Conclusion1The impact of dietary iron contents on the hematological indices of iron status,hippocampal iron status and other metal ions contents in male ratsDietary iron contents could significantly affect the hematological indices of ironstatus in male rats. Dietary iron deficiency could lead to nutritional iron deficiency,high dietary iron content could increase Hb, SI and lead iron overload in male rats.Dietary iron contents could not change NHI, sodium, magnesium, potassium,manganese and zinc contents in the hippocampus of male rats. Dietary iron deficiencycould increase copper contents in the hippocampus.2The impact of long-term exercise on the hematological indices of iron status,hippocampal iron status and other metal ions contents in male rats with differentdietary iron contentsIn the case of dietary iron deficiency, long-term exercise could further decreasethe hematological indices of iron status. In the case of dietary iron sufficiency andhigh dietary iron content, long-term exercise could not change the hematologicalindices of iron status, but increase RDW in male rats. With nutritional iron deficiency,sports people should improve dietary iron content or iron supplement.In the case of dietary iron sufficiency and high dietary iron content, long-termexercise could not change the hippocampus iron storage levels. Long-term exercisecould decrease the hippocampus iron storage in the case of dietary iron deficiency, and the mechanisms remain further investigation.Long-term exercise could not affect the contents of sodium, magnesium,potassium and manganese in the hippocampus. In the case of dietary iron deficiency,long-term exercise could increase zinc contents in the hippocampus. In the case ofdietary iron sufficiency, long-term exercise could increase copper contents in thehippocampus. Suggesting that with different dietary iron contents, long-term exercisecould lead to metal ions different change in the hippocampus. |
