Abstract/Summary:
Objective:To exam the expression of Her-2and Ki-67in different differen-tiationgastric carcinoma tissues. To analyze the correlation between both proteins and theirimpact on the clinicopathological features and the disease-free survival of gastriccarcinoma patients treated with postoperative adjuvant chemotherapy.Methods:Chose90cases of gastric carcinoma after radical surgery between January2008and July2008in Affiliated Hospital of Qinghai University.Patients receivedpostoperative adjuvant chemotherapy with fluorouracil and platinum majored regimenbut did not receive any preoperative treatments.The immunohistochemical stainingwas used to detecte the expression of Her-2and Ki-67,comprehensive analysis wasCarried out through combining with the clinicopathological features. To analyze thecorrelation between the expression of independent or co-expression in both proteinsand clinicopathological features.To Analyze the impact of expression of both proteinson the disease-free survival time after postoperative adjuvant chemotherapy..Results:1. The positive expression rate of Her-2in90cases of gastric carcinomawas39%while Ki-67was58%.No difference was observed in their expression onvarious differentiation of gastric carcinoma tissues.2. The expression of Her-2and Ki-67was positively correlated with TNMstage, depth of invasion and lymph node metastasis(P <0.05),had no correlationwith age and gender.3. There was a positive correlation between the expression of Her-2andKi-67(r=0.313,P<0.05),which was statistically significant.4. Her-2positive patients had significantly lower disease-free survival timethan that of Her-2negative patients; Ki-67positive patients had significantly lowerdisease-free survival time than that of Ki-67negative patients. Co-positive patients had the lowest disease-free survival time while the co-negative patients had thelongest (P<0.01).Conclusion:1. The expression of Her-2and Ki-67was positively correlated withTNM stage, depth of invasion and lymph node metastasis; there was a positiverelationship between the expression of Her-2and Ki-67.2.Results from the survivalanalysis showed that the patients with Her-2or Ki-67positive expression had lowerdisease-free survival time than patients with negative expression. Co-positive patientshad the lowest disease-free survival time while the co-negative patients had thelongest.Maybe the negative or co-negative patients of Her-2and Ki-67were moresensitive to fluorouracil and platinum combined regimen. Thus we think both Her-2and Ki-67play an important role in the metastasis of gastric carcinoma.Joint detectionmay be useful for the clinical assessment of tumor biological behavior and guidingpostoperative adjuvant chemotherapy regimen selection.
