| Objective:In this paper, using serum pharmacologic method, the Cs-4detection and evaluationon SMMC-7721cell proliferation, apoptosis and VEGF, MMP2expression effect.Methods:20body weight180~220g male SD rats were randomly divided into four groups:blank group and Cs-4low, medium and high dose groups. control group were treatedwith distilled water, Cs-4low, middle and high dose group, were treated with38.5mg/mL,77mg/mL,154mg/mLCs-4suspension, according to the1mL100g-1body weight orally,2times every day; intragastric administration of3D, preparationcontaining serum. The intervention serum Cs-4in vitro cultured SMMC-7721humanhepatocellular carcinoma cell line48h, the expression of MTT method, flowcytometry, ELISA detection of SMMC-7721human hepatocellular carcinoma cellproliferation, apoptosis and VEGF, MMP2.Results:TT results suggest: the proliferation of SMMC-7721cells was significantly inhibited(Cs-4in serum compared with the blank control group, P <0.05), in a dose-dependentmanner (Cs-4of different doses of group P <0.05). Find the cell apoptosis wasdetected by flow cytometry: low Cs-4serum, apoptosis, cells in high dose group were13.9%,23.5%,24.9%, were higher than those in the blank group (P <0.05). ByELISA detection of Cs-4in serum: low, middle and high dose group, theconcentration of VEGF in supernatant were respectively443.5,436.9,411pg/mL,lower than that in control group (P <0.05); Cs-4low, middle and high dose group, theconcentration of MMP2in supernatant were respectively14.7,14.3,14.2ng/mL, nodifference with the blank group (P>0.05).Conclusions:The serum containing Cs-4has inhibition on proliferation of hepatoma cells,inhibit the strength increases with the increase of drug concentration; Cs-4containing serum could increase the apoptosis of SMMC-7721cells cultured in vitro rate;inhibition of VEGF expression, suggesting that Cs-4has anti metastasis effect; alsofound that Cs-4containing serum on SMMC-7721cell MMP2expression had nosignificant inhibitory effect. |
