Objective: The aim of this study was to evaluate the inhibitory effects of GinsenosideRh1on proliferation, invasion and metastasis ability of colon cancer. Its potentialmolecular mechanism was discussed.Method:cells were divided into five groups,control, PMA, PMA+30μΜ Rh1, PMA+100μM Rh1, PMA+300μM Rh1.The survival ratio of colon cells treated by differentconcentrations of Rh1were measured by MTT assay exclusion test The invade andmigrate ability of colon cancer cells were measured by scratch wound assay andartificial basilar membrane matrigel respectively.The expression of MMP-9of coloncancer cells was measured by ELISA kit. RealTime-PCR and Western blot were usedto evaluate the mRNA and protein expression of MMP-9Results:PMA significantly enhanced colon cancer cell proliferation,Invasion andmigration, whereas Rh1suppressed PMA stimulation of cell proliferation, Invasionand migration. Rh1suppressed PMA stimulation of cells MMP-9protein expressionof and MMP-9mRNA expression.Conclusion: All the results demonstrate that Rh1plays an important role in theanti-metastasis and anti-Invasion of colon cancer possibly by inhibit the activity ofMMP-9.
