Objective:To detect methylation status of the promoter region of p15、syk genein rectal cancer,the relationship between the aberrant methylation of p15、Syk geneand postoperative recurrence and metastasis after resection of colon cancer wasanalyzed.Methods: By using nested polymerase chain reaction(Nested PCR)andmethylation-specific polymerase chain reaction technique (MSP), methylation ofp15、Syk promoter region from55rectal cancer patients (tumor tissues, adjacentnormal tissues) were detected. To investigate the relationship between methylationstatus of the promoter region of p15、Syk gene and the postoperative recurrence andmetastasisResults:The frequency of methylation of p15、Syk promoter region was56.4%(31/55)and36.4%(20/55)in rectal cancer.In contrast, The frequency of methylationof p15, Syk promoter region was16.4%(9/55)and0%(0/55)in adjacent normaltissues. the rate of methylation of p15、Syk promoter in rectal cancer was higher thanthat of adjacent normal tissues(P<0.05). The frequency of postoperative recurrenceand metastasis in31cases with p15promoter methylation was32.2%(10/31) and thatof Syk was in35.0%(7/20). the promoter methylation of p15and Syk are associatedwith differentiation,TNM(P<0.05). The number of the promoter methylation of p15was31,in which the the number of the promoter methylation of Syk is17at the sametime.Conclusion:(1)p15、Syk gene promoter methylation may play an importantrole in occurrence and progression of rectal cancer(2)Detecting the expression ofp15and Syk gene in rectal carcinoma could improve the early diagnosis of rectalcancer and evaluate the prognosis.
