Objective:To analysis the relevant factors of malignant change of hydatidiformmole (HM),through epidemic learn investigation from medical history and related factors fromdiagnosis and treatment.To analysis the expression of imprinted gene H19and IGF2in hydatidiform mole tissue through RT-PCR-RFLP technology in gene molecularlevel.Detectthe gene expression differences between the malignancy group and notmalignancy group, provides reference for reasonable treatment and the prognosis tohydatidiform mole patients.Methods:Collect30cases of HM diagnosed in the curettage for the first time, andanalysis the differential gene expression of imprinted gene H19and IGF2betweenhydatidiform mole and malignancy hydatidiform mole, through RT-PCR-RFLPtechnology on gene molecular level. Then follow-up the40patients.Through analysisthe history of epidemiological and clinical data in the process of investigation, foundthe related factors of HM malignant change.1. In this experiment there was no correlation has be found that hydatidiform molenumber of evil change with age, pregnancy, medicine flow history, birth control pillsor hormone therapy history, history of vaginal bleeding, Hyperthyroidismperformance, history of infertility treatment, pesticide exposure history, HCG sizebefore evacuation, size of the uterus, the degree of postoperative pathology, sertolicell hyperplasia about hydatidiform mole and Malignant transformation ofhydatidiform mole, there was no statistically significant difference (P>0.05). Historyof spontaneous abortion and HCG hasn’t be Log down after evacuation were relatedwith Hydatidiform mole malignant transformation,there was statistically significantdifference (P<0.05).2.30cases of hydatidiform mole tissues, there were8cases of H19heterozygote,there were12cases of IGF2heterozygote, there were3cases of H19and IGF2heterozygote, State of H19gene detection results:4specimens of hydatidiform mole for Double allelic gene expression,4specimens of hydatidiform mole single allelicgene expression;12specimens of hydatidiform mole IGF2gene detecting state:5specimens of hydatidiform mole for double allelic gene expression,7for single allelicgene expression. The malignant hydatidiform mole occurred in4cases,2cases arehomozygous,1case is H19heterozygote,1case is IGF2heterozygote, both H19andIGF2gene were double allelic gene expressioned.Conclusions:1. In this experimental spontaneous abortion and HCG is not a logarithmic dropafter evacuation24hours are associated with malignant change hydatidiform mole2. Expression of the Mark gene H19and mark IGF2have no relationshipbetween Malignant transformation of hydatidiform mole.
