Objective:To explore AgNPs-coated sutures reducing inflammatory cells infiltration andpromoting tissue remodeling around sutures using an running intradermic suturemodel in mice.Methodolgies:1、AgNPs was coated on absorbable sutures(5-0, VICRYL)by layer-by-layerdeposition approach and observed the morphology and distribution of AgNPs onsurface of sutures by Scanning Electron Microscopy (SEM).2、Make a longitudinal incision on dorsal skin in mice. The respective sutures(VICRYL、 VICRYL Plus、 AgNPs) were then used for intra-cutaneous serialsaturation on dorsal skin in mice.3、The intra-cutaneous serial saturation sites were harvested to investigate thedegree of tissue inflammation about neutrophils,macrophages,TNF-α,as well as thesituation of collagen deposition.Results:1、SEM observation indicated AgNPs could be immobilized and distributedon suture surface evenly. AgNPs-coated suture had the best in vivo anti-bacterial efficacy when compared with other groups.2、Immunohistochemistry results showed significantly less inflammatory cell infiltration (macrophage, neutrophil)(P<0.05) and pro-inflammatory protein expression (TNF-α)(P<0.05).3、Masson staining showed the better collagen deposition in the intra-cutaneousserial saturation tissue in the AgNPs-coated suture group,which was similar to thenormal skin tissue.Conclusion:1、AgNPs-coated sutures can reduce inflammatory cells infiltration andpromote tissue remodeling around AgNPs-coated sutures. 2、 AgNPs-coated sutures have potential to promote ideal wound healing aftercoated.
