The Relationship Between GSTP1and Smoking Related Lung Adenocarcinoma

Aim:1. To investigate the expression of GSTP1and EGFR in human lungadenocarcinoma with long smoking history(over30years) or second-smoking history(for working five days a week over45years in smoking places or visiting smokingplaces for an average of13minutes a day over60years), and discuss the relationshipbetween GSTP1and EGFR.2. To investigate the gene and protein expression of GSTP1in HBE, A549,NCI-H23, NCI-1299which were treated by CSE. To study the proliferation of HBE,A549, NCI-H23, NCI-1299which were treated by CSE.Methods:1. Collected human lung adenocarcinoma tissues diagnosed in the secondaffiliated hospital of Nanchang University between2012-1013. The gene expressionof GSTP1and EGFR in these tissues of were evaluated by Real-Time quantiativeReverse Transcription PCR.2. The gene expression of GSTP1and EGFR in HBE, A549, NCI-H23,NCI-1299, treated by CSE was evaluated by Real-Time quantiative ReverseTranscription PCR.3. The proliferation of HBE, A549, NCI-H23, NCI-1299treated by CSE wasevaluated by MTT assay.Results:1. Among28cases tissues, the gene expression of GSTP1and EGFR downregulated in10lung adenocarcinoma tissues，meanwhile13cases up regulated.2. Compared with HBE, the down regulation of GSTP1and EGFR wasevaluated, and there was no significant difference between the expression of GSTP1and EGFR in the same cells. The up regulation of GSTP1and EGFR was evaluated inNCI-H23treated by CSE, meanwhile the down regulation of GSTP1and EGFR wasevaluated in A549and NCI-1299treated by CSE. 3. The up regulation of the protein expression of GSTP1was evaluated in HBE,A549, NCI-H23, NCI-1299, after treated by CSE.4. The IC50of HBE, A549, NCI-H23, NCI-1299treated by CSE, respectively，is3%，5%，3%，8%.Conclusion:1. The up regulation of GSTP1and EGFR was evaluated in cases had longsmoking history(over30years) or second-smoking history(for working five days aweek over45years in smoking places or visiting smoking places for an average of13minutes a day over60years). GSTP1may participate the occur of smoking relatedlung adenocarcinoma.2. The up regulation of the protein and gene expression of GSTP1was evaluatedin HBE treated by CSE. GSTP1may participate the elimination of carcinogen intobacco.3. The up regulation of the protein and gene expression of GSTP1and NCI-H23was evaluated in HBE, meanwhile the down regulation of the gene expression ofGSTP1in A549and NCI-1299and the up regulation of the protein expression ofGSTP1in A549and NCI-1299. The significant difference between HBE, A549,NCI-H23, NCI-1299, GSTP1inclined more efficiency in HBE.4. Compared with HBE, A549, NCI-H23, NCI-1299have more resilience intobacco.
