Objective: Analysis of IFN-γ gene region of qinghai Tibetan crowd2polymorphism loci and the relationship between the susceptibility to chronic hepatitis b,select relevant major sites, for the region of qinghai Tibetan population of HBVsusceptibility risk prediction lay the foundation.To provide new theory for clinicaldiagnosis and prevention of hepatitis b and basis.Methods:(1) the object of study: in250cases of chronic hepatitis b patients tibetan in qinghai area (case group) and134cases of healthy subjects (control group) as the research object.(2) gene polymorphismanalysis: peripheral blood DNA extracted with the use of blood genome smallextraction kit, use of flight mass spectrometry analysis of gamma interferon gene+874A/A、+874A/T、+874T/T、+2109C/C、+2109C/T、+2109T/T genotype.(3) data analysis: calculate each point genotype and allele frequency, comparison ofeach genotype differences between groups, and Hardy-Weinberg equilibriumalignment inspection, if p <0.05, the application of logistic multivariate regressionanalysis of the genotypes and the correlation of susceptible to chronic hepatitis b, if p>0.05, gene polymorphism and the susceptibility of hepatitis b has nothing to do.Results(1)+2109loci in the frequency distribution between the case group and the controlgroup.①+2109C/C in the case group and the control group were10.5%and14.3%respectively, the statistical analysis results show that the frequency distributionbetween groups did not see significant difference.②+2109C/T in the case group andthe control group were47.3%and43.7%respectively, the statistical analysis resultsshow that the frequency distribution between groups did not see significant difference.③+2109T/T in the case group and the control group were42.2%and42.1%respectively, the statistical analysis results show that the frequency distribution between groups did not see significant difference.(2)+874loci in the frequencydistribution between the case group and the control group.①+874A/A in the casegroup and the control group were3.1%and2.4%respectively, the statistical analysisresults show that the frequency distribution between each group did not see significantdifference.②+874a/T in the case group and the control group were30.6%and19.8%respectively, the statistical analysis results show that the frequency distributionbetween groups did not see significant difference.③+874T/T in the case group and thecontrol group were66.3%and77.8%respectively, the statistical analysis results showthat the frequency distribution between groups did not see significant difference.(3)+874T allele gene frequencies in the case group and the control group were81.6%and87.7%respectively, the statistical analysis to carry the frequency of allele T risesuggests that the lower the susceptibility of second liver.Conclusion(1) IFN-γ+874polymorphic loci and the region of qinghai Tibetan population susceptibility to HBVinfection may has no any relation.(2) IFN-γ+2109polymorphic loci and the region ofqinghai Tibetan population susceptibility to HBV infection may has no any relation.(3)IFN-γallele T+874May be played a role of qinghai tietan population susceptibility toHBVprotective genes.
