In most cases, apoptosis are non-immunogenic or even immunosuppressive. However,recently, Studies have shown that some chemotherapy drugs killed tumor cell can induce theanti-tumor response in patients. our previous study has indicated that clostridium difficiletoxin B, secreted by clostridium difficile, can elicit the immunogenic apoptosis of CT26tumorcell.It was reported that HSPs exposure or secretion is a important feature of immunogenicapoptosis. Here, we explore whether there are the exposure or secretion of HSPs in thissystem. And if the answer is yes, do HSPs affect the immunogenity of TcdB treated CT26andhow it affects the Immune response.Finally, we found that there are the secretion of HSP70and HSP90but no the exposureof them in TcdB induced immunogenic apoptosis system. Blockade or inhibition of HSPsinfluences the immunogenicity of dead CT26in mice. And they affect the anti-tumor immuneresponse because of its effect on DC mature. Furthermore, our findings also show that HSP70which secreted from treated CT26cell can interact with TLR4, a recepter on the surface ofDC. This provide a clue to find the mode which HSPs promote DC mature.These data have some significance for the development of immunogenic apoptosismechanism.It’s also a surport for the exploration of vaccine based on TcdB.
