Objective:To investigate the protection of the total lignans from Tibetan MedicinalHerpetospermum Seeds on experimental liver injury induced by CCl4and ConA and furtherto discuss protective mechanism against immunologic liver injury induced by ConA.Methods:Establish the model of acute liver injury by intraperitoneal injection of0.1%CCl4olive oil solution20ml/kg.After16h fasting,take blood samples and detect alanineaminotransferase (ALT),aspartate aminotransferase (AST) and alkaline phosphatase(ALP)activities in serum.Meantime,take a portion of the liver into the pathology observation.Inject with ConA (20mg/kg) from tail vein in order to induce immune hepaticinjury.Animals were sacrificed at the8th hour and,blood samples and liver wascollected.Test index of liver and investigate the effect of different doses of the totallignans from Tibetan Medicinal Herpetospermum Seeds on ALT,AST,ALP,NO,NOS inserum and SOD,MDA,GSH-Px in liver tissue of immunologic liver injury mice.HEstaining was performed for observing the pathological chang of the liver tissue.Necrosisfactor alpha (TNF-α),interferon gamma (IFN-γ),interleukin-4(IL-4),interleukin-10(IL-10),a nuclear transcription factor (NF-κB) were tested with immunohistochemicalmethod.Results:The total lignans from Tibetan Medicinal Herpetospermum Seeds can reduceALT,AST,ALP in serum of liver injury mice induced by CCl4.Meantime,They can re-duce ALT,AST,ALP activities and weaken overexpression of NO,NOS in serumof immunological liver injury mice induced by ConA.MDA were lowered signific-antly and the activities of SOD and GSH-Px were markedly increased in liverhomogenate.The expression of TNF-α,IFN-γ,IL-4and NF-κB were inhibited.Theanti-inflammatory cytokine interleukin-10(IL-10) expression was promoted. Based on the reasons above,the total Lignans from Tibetan MedicinalHerpetospermum Seeds play a role in protecting the liver.Pathological results alsoshow that the total Lignans from Tibetan Medicinal Herpetospermum Seeds can redu-ce liver disease and improve CCl4and ConA liver tissue damage.Conclusion:The total Lignans from Tibetan Medicinal Herpetospermum Seeds have acertain protective effect on CCl4-induced chemical liver injury and ConA-induced imm-unological liver injury in mice.The total Lignans from Tibetan Medicinal Herpetos perm-um Seeds on ConA-induced liver injury may be related to enhancement of radical scaveng-ing capacity and the body’s anti-lipid peroxidation, reduction of inflammatory cells aggreg-ation in areas of inflammation and tissue damage, inhibition of inflammatory cytokine tu-mor necrosis factor-α (TNF-α),interferon-γ (IFN-γ),interleukin-4(IL-4),the nuclear tra-nscription factor (NF-κB) release,and promotion of the anti-inflammatory cytokine interl-eukin-10(IL-10) expression.
