Background: Endometrial stem cell hypothesis proposed that when menstrual blood reflux,endometrial stem cells cultivation to the abdominal cavity is the key of endometriosisformation, but there is no experimental evidence.Objective: Explore the existence of endometrial stem cells in mice. Experimentalendometriosis was established by transplanting endometrial cell or tissue from greenfluorescent protein (GFP) mice to the peritoneal cavity of ICR mice.Methods:1. Isolate endometrial cells to identify and quantify the live SP cells in murine endometriumwith the Hoechst33342-SP method by flowcytometry.2. Transplanting endometrial cell or tissue from green fluorescent protein (GFP) mice to theperitoneal cavity injured area of ICR mice. Experimental groups:1) endometrial cellstransplanted group2) endometrial cells and bone marrow mesenchymal cell co-culturetransplanted group3) digested endometrial tissue transplanted group4) pelvic injury3weeks before transplantation of digested endometrial tissue group.Continue feeding4-6weeks, then we observed the endometriosis lesions directly byfluorescence microscopy, and under the microscope we did HE staining and Anti-GFPantibody immunohistochmical staining.Result:1. Fresh digested single cell staining with Hoechst33342can detect SP cells byflow-cytometry; the percentage of SP cells is1.25-2.29%.2.The mice transplanted digested endometrial tissue developed endometriosislesions.Therefore, Has a history of recent injury mouse model have a higher growth ratethan fresh injury mouse model, respectively100%(12/12) VS37.5%(3/8), Anti-GFPantibody Immunohistochmical staining positive could be found in growth lesions.Conclusion: Mouse endometrial cells contain SP cells.Transplantation of digestedendometrial tissue is easier to form endometriosis lesions than transplantation of endometrialcells, the history of recent injury increases the success rate of endometrial xenograft model.
