【Objective】 To investigate the association between single nucleotide polymorph-hisms in the HLA-DP, DQ genes and the outcomes of chronic hepatitis B virusinfection in Fujian population.【Methods】204Health subjects,255Clearance subjects,204asymptomatic HBVcarrier,136chronic hepatitis B,68liver cirrhosis and heptocellular carcinoma wereenrolled in this study. Genotyping of rs3077, rs9277535, rs2647050were performedby Polymerase Chain Reaction-Sequence Specific Primers (PCR-SSP).【Results】Distribution of genotype of the rs3077, rs9277535, rs2647050locus werein accordance with Hardy-Weinberg equilibrium in Health subjects, Clearancesubjects and asymptomatic HBV carrier. By using Health subjects and Clearancesubjects as control group, rs3077A and rs9277535A were significantly associated withchronic HBV infection under additive and dominant model (P<0.05), other thanrs2647050(P>0.05). meanwhile, haplotype GGA, AGA, AAA served as protectivefactors against chronic HBV infection (P<0.05). By using asymptomatic HBV carrieras control group, compared to chronic hepatitis B group, liver cirrhosis group andheptocellular carcinoma group, our study showed that these3SNP and haplotype noassociated with clinical outcomes(P>0.05).【Conclusion】 HLA-DP gene polymorphisms were strongly associated with chronicHBV infection, and the presence of the A allele at rs3077and rs9277535of theHLA-DP gene may decreased the risk for chronic HBV infection. But HLA-DP andDQ genes polymorphisms no associated with the outcomes of chronic hepatitis Bvirus infection. 【Objective】 To assess the rigorous relationship between HLA-DP polymorphisms(rs3077, rs9277535) and hepatocellular carcinoma by mean of Meta-analysis.【Methods】A Meta-analysis was conducted with the data from three papers relevantto hepatocellular carcinoma, following strict selection. The pooled odds ratios (OR)with95%confidence intervals (CI) were calculated using fixed-or random-effectsmodel when appropriate. Funnel plot and Egger’s test was used to measurepublication bias.【Results】Our analysis suggested that the presence of the A allele at rs3077of theHLA-DP gene may decreased the risk for hepatocellular carcinoma (OR=0.83,95%CI:0.74~0.93) under the dominant model, but rs9277535no associated with thehepatocellular carcinoma (AA+AG vs GG:OR=0.95,95%CI:0.85~1.06).【Conclusion】By Meta-analysis, out study confirmed that the presence of the Aallele at rs3077of the HLA-DP gene may decreased the risk for hepatocellularcarcinoma.
