Objective: To observe the effect on the human umbilical vein endothelial cells(EA.hy926) structure and function induced by human tumor necrosis factor-a (TNF-α),and to explore its mechanism.Methods: The EA.hy926cells were treated with1,10,100ng/ml TNF-α for24hoursrespectively, or100ng/ml TNF-α for3,8,12,24h. The protein expression of vasodilatorstimulated phosphoprotein (VASP) was dectected by Western blotting, while VASPmRNA expression was by real-time quantitative PCR (Real-time PCR). Cells apoptosiswas analyzed by flow cytometry. The ultrastructural changes of the cells were observedby transmission electron microscopic.Results: TNF-α decreased endothelial cell VASP protein and mRNA expression in adose-and time-dependent manner. In the condition of prolonging exposure time effectsor increasing concentrations of TNF-α, the damage level of cells got worse, and the ratioof apoptosis was increased.Conclusion: TNF-α increased the permeability of vascular endothelial cells bydestroying the endothelial structure and function in a dose-and time-dependent manner.
