ObjectiveTo investigate the relationship between the expressions of MMP-9, TIMP-2, nm23, CD44, E-Cadherin, Integrin and pathological factors and lymph node metastasis in colorectal carcinoma. And to explore factors related to lymph node metastasis of colorectal carcinoma.MethodsThe medical records, paraffin-embedded specimens and pathological sections of105patients with colorectal carcinoma were collected. The tissue samples were used to make manual tissue microarrays. The immunohistochemistry method was used to detect the expressions of MMP-9, TIMP-2, nm23, CD44, E-Cadherin, Integrin in colorectal carcinoma with and without lymph node metastasis.Results(1) The negative correlation between expression of TIMP-2and lymph node metastasis of colorectal carcinoma was observed (r=-0.211,P=0.033).And the negative correlation between expression of E-Cadherin and lymph node metastasis was observed (r=-0.283, P=0.004). There was a close relationship between E-Cadherin and lymph node metastasis of colorectal carcinoma.(2) We recorded a statistically significant positive correlation between expression of MMP-9, and TIMP-2; positive correlation between E-Cadherin and IntegrinP1; positive correlation between TIMP-2and E-Cadherin; and the same as nm23and MMP-9, TIMP-2, IntegrinP1.(3) The expression of TIMP-2was associated with differentiation (P=0.035). The expression of E-Cadherin was associated with the histological type (P=0.001), differentiation (P=0.007). And the expression of Integrinβ1was associated with the histological type (P=0.044).(4) There was a connection between differentiation and lymph node metastasis of colorectal carcinoma (P=0.001).(5) Risk factors of lymph node metastasis in colorectal carcinoma were differentiation, invasion depth, MMP-9, CD44; protective factors were TIMP-2、nm23、E-Cadherin、Integrinβ1. Differentiation and E-Cadherin were closely associated with lymph node metastasis in colorectal carcinoma.Conclusion The expressions of TIMP-2and E-Cadherin play important roles in predicting the lymph node metastatic potential and prognosis in patients with colorectal carcinoma.
