Objective: To investigate the expressions of β-cat,CyclinD1and Ki-67protein andHPV16/18,31/33DNA in invasive squamous cell carcinoma of cervix(ISCC), and analysetheir significance in the occurrence and development of ISCC.Methods: The expressions of β-cat, CyclinD1and Ki-67protein in73cases of ISCC,30cases of high-grade squamous intraepithelial lesions (HSIL),32cases of low gradesquamous intraepithelial lesion (LSIL), and21cases of normal cervical squamousepithelium (NCE) were explored by tissue microarray and two-step immunohistochemistrymethods. The expressions of HPV16/18,31/33DNA were detected by in situ hybridization.The results were analyzed by SPSS13.0statistical software.Rerults:1. The results of in situ hybridization showed that the positive rates of HPV16/18expressions in ISCC、HSIL、LSIL and NCE were65.75%(48/73)、46.47%(14/30)、9.38%(3/32)and0%(0/21),respectively. The positive rates of HPV31/33were50.68%(37/73)、30%(9/30)、12.50%(4/32)and0%(0/21),respectively.2.The expressions of HPV16/18,31/33DNA showed a statistically significant positivedifference in cervical lesions in the four groups (p<0.01); Intravascular tumor thrombuswere more likely detected in HPV16/18infection cases than non-infectionones(OR=3.875), HPV31/33-positive patients during clinical period were more likely toprogress to higher level.3.The different expressions of β-cat, CyclinD1and Ki67proteins in the four groups were statistically significant in the overall (p<0.01); β-cat demonstrated heterogeneity ofexpression in ISCC、HSILand LSIL, the HSIL group in the β--cat heterogeneity ofexpression beyond the normal group (p<0.001).4.Ki67-positive expression in low grade intraepithelial lesion group wassignificantly higher than that in the normal group (p=0.003).5.CyclinD1-positive expression between invasive squamous cell carcinoma and thehigh-level intraepithelial lesions was statistically significant (p=0.008).6.The pathological stage of squamous cell carcinoma of the cervix increased with age(OR=1.063).Conclusions:1.HPV16/18and HPV31/33infections are closely related to the occurrence ofsquamous cell carcinoma of cervix and its development.2. HPV16/18and HPV31/33infections are the risk factors of intravascular tumorthrombus and clinical stage, respectively. This indicate that both of them are bound up tothe clinical progression and poor prognosis of squamous cell carcinoma of cervix.3. The abnormal expression of cyclin D1is concerned with the occurrence ofsquamous cell carcinoma of cervix, and the over-expression of it manifests on the earlystage of cancerization of cervix.Ki67can be conducted as the objective reference criterionfor diagnosis of iintraepithelial lesions of cervix..4. Ki67and β-cat can be regarded as cervical intraepithelial lesionsespecially intraepithelial lesion level evaluation indicators.
