Objective To optimize the therapeutic dose and time window of picrosede Ⅱ in cerebral ischemic injury in rats by orthogonal test.Methods The forebrain ischemia models were established by bilateral common carotid artery occlusion (BCCAO) methods. The successful models were randomly grouped according to orthogonal experimental design and treated by injecting picroside Ⅱ intraperitonenally at different ischemic time with different dose. The concentration of hydrogenperoxide (H2O2) and the activity of catalase (CAT) in serum and brain tissue were determined by chemiluminescence immunoassay.Result:The optimized composition of the therapeutic dose and time window of picroside Ⅱ in cerebral ischemic injury was (1) ischemia1.5h with10mg/kg and ischemia1.5h with20mg/kg body weight according to the concentrations of H2O2in serum and brain tissue respectively.(2) ischemia1.5h with20mg/kg and ischemia1.5h with10mg/kg body weight according to the activities of CAT in serum and brain tissue respectively.Conclusion:Combined evaluation from the principle of lowest therapeutic dose with longest tome window, the optimized composition of the therapeutic dose and time window of picroside Ⅱ in cerebral ischemic injury is10-20mg/kg body weight at ischemia1.5h.
