Objective:To investigate the influence of78KDa glucose-regulated proteinson neutrophil apoptosis in rat with severe acute pancreatitis induced bysodium taurocholate.Methods:Fifty-four male SD rats were randomized into sham group(SO)(n=24) and severe acute pancreatitis(SAP) group(n=30). The SAP wasinduced by injections of5%sodium taurocholate via retrogrzdecholangiopancreatography. The sham group only open abdomen and flippancreas, then suture. The animals were sacrificed at3h,6h and12h later,blood samples were collected from femoral vein to measure serum amylase,PMNs were separated and the rate of apoptosis of PMNs was tesed by flowcytometry, extract the proteins of PMN to test the expression of78KDaglucose-regulated protein and Mcl-1protein via western-blot and observe thepathological changes of the pancreas by HE staining.Results: The density of serum amylase of the severe acute pancreatitis groupwere higher than that of control group (P<0.01) and the SAP group wereincreased gradually at3h,6h and12h (P<0.01). Pathlogical HE stainingshowed that there are numerous inflammatory cells and erythrocyte infiltrated.The rate of apoptosis of PMN was decreased remarkably in the SAP group than in the SO group, and the decrease became more obvious over time(P<0.01). The western-blot revealed that the expression of GRP78and mcl-1in SAP group increased significantly compared with that in SO group. Therewas a negative correlation between the apoptosis rate of PMN and theexpression of GRP78and Mcl-1in SAP rats (r=-0.836,P<0.01)、(r=-0.895,P<0.01).Conclusion: The rate of apoptosis of PMN declined over time in rat、severeacute pancreatitis, and the expression of78KDa glucose-regulated proteinand Mcl-1protein were remarkably increased, it may be related to theincreased GRP78generated by stress by PI3K/Akt pathway activated theMcl-1, and finally plays an important role in the neutrophil apoptosis.
