Background and purpose Major histocompatibility complex Ⅱ (MHC Ⅱ) is atransmembrane glycoprotein that its main biological effects associated with tumorAntigen processing and presentation. Current research has shown that, MHCⅡinvolvedin the occurrence and development of multiple tumor. This experiment throughdetection of diffuse large B-cell Lymphoma of major histocompatibility complex MHCⅡmolecule expression and explore its clinical pathological significance.Methods Using Immunological Histological Chemistry (IHC) technique EV two-stepon123patients with diffuse large B-cell lymphoma immunophenotyping, and detect theexpression of30cases of RH tissues and123cases of DLBCL tissues in MHCⅡmolecules. While applying the Chromogenic In Stiu Hybridization (CISH) to detectthe expression of MHC II of84patients with DLBCL and30cases of RH tissues,inorder to analyze the relationship between MHC II protein and mRNA expression andclinicopathological characteristics and prognosis in DLBCL patients.Results1. Using CD10,BCL-6and MUM1to immunophenotyping the123casesDLBCL tissues. There are71cases (57.7%) of ABC type DLBCL and52cases (42.2%)of GCB type DLBCL.2.The IHC data showed that the positive rates of MHCⅡprotein and mRNA were49.5%(61/123),46.43%(39/84) in DLBCL tissues andsignificantly lower than it in RH tissues, which is86.7%(26/30),90.00%(3/30). Thepositive rates of MHCⅡwere38%(27/71) for ABC type DLBCL and significantly lower than it in GCB type DLBCL, which is65.3%(34/52).3. The expression ofMHCⅡwere significantly lower in more extranodal involvement of multiple lesions,higher ECOG scores and High-risk group of international prognostic index (P<0.05).But the expression of MHC protein and mRNA is not associated with otherclinicopathologic characters (P>0.05).4. In ABC type DLBCL, the expression ofMHCⅡ has inverse correlation with the expression of MUM1(P<0.01,r=-2.42).Conclusions The results of this study indicated that low expression of MHCⅡmolecules was present in diffuse large B-cell lymphoma. Its abnormal expression mayplay an important role in the progress and infiltration of diffuse large B-cell lymphoma.The loss of its expression prompted a poor prognosis in DLBCL patients. Overall, thedata from our study show that MHCⅡ may play a role in DLBCL pathogenesis.
