ObjectiveStudies have found that lung cancer’s incident and metastatic are closely related to STK33. But it unknown the mechanism that STK33how to act on biological characteristics of lung cancer. Tumor cells’unlimited proliferation and metastasis are an important characteristic. p53and CDC2associated with a variety of tumors, they can regulate cell mitosis and apoptosis. CXCR4is closely related to metastasis of tumor cells. This paper studies whether STK33effect on p53、CDC2、CXCR4in lung cancer. Explore STK33in lung cancer how to impact on the development and prognosis in lung cancer.MethodsUsing the established stable with low metastatic lung cancer cell lines NL9980and L9981cells with high metastatic lung cancer as the experimental group to conduct cell culture and plasmid transfection.Then constructing the expression experimental control groups of NL9980(+) and L9981(-) cells.Using real-time quantitative RT-PCR and western-blot methods to detect the expression of STK33in lung cancer cells whether change the expression of cell cycle-related proteins (p53and CDC2) and surface proteins (CXCR4)’s expression levels.Results(1)STK33in low metastatic lung cancer cells NL9980mRNA below the L9981lung cancer cells with high metastatic (P<0.001)(2) After STK33is highly expressed in the low metastatic lung cancer cells NL9980, p53of NL9980(+)’s mRNA and protein were lower, L9981highly metastatic lung cancer cells after low expression STK33, p53of L9981(-)’s mRNA and protein were increased.(3) After STK33is highly expressed in the low metastatic lung cancer cells NL9980, CDC2of NL9980(+)’s mRNA and protein were increased, L9981highly metastatic lung cancer cells after low expression STK33, CDC2of L9981(-)’s mRNA and protein were lower.(4) After STK33is highly expressed in the low metastatic lung cancer cells NL9980, CXCR4of NL9980(+)’s mRNA and protein were increased, L9981highly metastatic lung cancer cells after low expression STK33, CXCR4of L9981(-)’s mRNA and protein were lower.Conclusions(1) STK33expression levels in highly metastatic lung cancer cells are higher than the low metastatic lung cancer cells. STK33may prompt the degree of lung cancer cells and cell malignant.(2) STK33expression level changes in lung cancer cells have a close relationship with expression of cell cycle proteins p53and CDC2. May be STK33by affecting cell cycle regulation to involve in the development of lung cancer.(3) STK33expression level changes in lung cancer cells have a close relationship with expression of membrane surface protein CXCR4. STK33may affect metastasis of lung cancer by acting on the membrane surface protein of CXCR4.
