Objection:To study the correlation of tumor microenvironment and colorectal cancer through comparatively analyzing the molecular feature of ECM in early colorectal cancer at different syndromes and distance tissue from tumor. Accordingly, the research provide powerful clue to prevent and cure colorectal cancer and search new anti-cancer targets. Meanwhile, it offers effective biological foundation for syndrome differentiation of TCM and correct drug usage.Methods:The methods of immunohistochemistry and Western Blot were used to detect E-cad、HYAL1and ColI expression in twenty colon moderately differentiated adenocarcinoma specimens (Dampness-Heat syndrome and the syndrome of yang deficiency of spleen and kidney respectively ten cases), corresponding normal tissues (ten cm apart from tumor)、para-cancerous tissues (five cm apart from tumor and two cm apart from tumor), which were named as positionl,2and3. The method of HE staining observe the crypt structure of different position tissue. Reaserching the expression and distribution of related ECM, consequently obtaining conclution.Results:1.HE staining show:the crypt structure is different at different distance from tumor tissue.Position1:the crypt structure is normal. Cells show monolayer arrangement and nucleus and epithelial layer are normal. Position2:the crypt completely disappear or segment remaining.The crypt appear broad and the crypt lumen in these foci appear with an irregular lumen compared with the normal crypt. Cell nucleus appear larger, cell arrangement disorder and epithelial cell stratification. Position3:the crypt appear dysplasia and broad, irregular lumen, thickening epithelial, larger nucleus, nuclear hyperchromasia and epithelial cell stratification.2. Immunohistochemistry show:the expression and distribution of ECM(Col-1、HYAL1、E-cad) exist differences at different distance from tumor tissue. Col-1is found primarily in connective tissue of submucosa and epithelial layer, a little among crypt. The corrected optical density are1.8453±0.2586,2.1539±0.2353,1.9723±0.2091respectively in position one, two, three. HYAL1is found primarily in connective tissue of submucosa. Its optical density are1.5823±0.2686,1.8715±0.2016,1.8095±0.1970. E-cad is found primarily in cytomembrane and cytoplasm. Its optical density are2.0819±0.2347,1.8224±0.2384,1.7586±0.3511.3. Western Blot show:the expression levels of E-cad are respectively5.5635,1.9875,0.9210, reflecting a degression trend. The expression of Col-1and HYAL1are maximum in position two. Their expression levels are respectively1.3119,2.4604,0.8699,1.4635,2.5462,0.8646in three positions.4. Immunohistochemistry analysis show:the expression of ECM (E-cad、ColI、 HYAL1) exist difference to some extent at different syndromes. The expression of Dampness-Heat syndrome exceed the expression of the syndrome of yang deficiency of spleen and kidney. But they have no statistical significance. Conelusion:1.HE staining show:the lesion of crypt structure is more serious and ACF are more prominent and larger closer to the tumor.2. Immunohistochemistry and Western Blot show:the expression of E-cad reflects a degression trend in three positions. The expression of Col-1and HYA1are maximum in position2. Their expression trend are firstly ascend then degression. ECM and tumor microenvironment play a significant role in the occurrence of colorectal carcinoma.3. the expression of ECM (E-cad、ColI、HYAL1) exist difference to some extent at different syndromes. The expression of Dampness-Heat syndrome exceed the expression of the syndrome of yang deficiency of spleen and kidney. And the expression of Col-1and HYAL1and pathological stage of neoplasia display positive correlation.the expression of E-cadherin and pathological stage of neoplasia display negative correlation.
