Objective1To analyze the expression of Livin protein in different pathological types of gastric mucosa.2To analyze the relationship between the Livin Protein and the clinical features of gastric cancer.3To analyze the correlation between the Livin protein and the happening, development, TCM syndromeof gastric cancer.4To study the effects of Livin gene transfection on cell proliferation, apoptosis in gastric cancer cell line MG-803.Methods1Collect totally151cases which were treated in department of gastroenterology or department of general surgery of Guangzhou General Hospital of Guangzhou Military Command of PLA from2008to2012, including30cases of chronic gastritis,36cases of gastric ulcer,54cases of gastric cancer and31cases of cancer adjacent tissue. All of them were confirmed by pathological biopsy through endoscope or surgery and chemotherapy or radiation therapy was not applied before surgery.2Make the syndrome differentiation in Traditional Chinese Medicine for the gastric cancer patients by using the four diagnostic parameters.3Examine the expression of Livin protein by SP immunohistochemistry in totally151cases, and analyze the positive rate in each group. 4Build the eukaryotic recombinant plasmid Livin by liposome transfection of exogenous genes to Livin MGC-803gastric cancer cell lines. By using RT-PCR to detect the expression of Livin mRNA, MTT method to compare the cell growth curve, and analyzed the changes of cell by flow cytometry;5Analyze the results by statistics software SPSS13.0.Results1The positive rate of Livin expression in gastric cancer was significantly higher compared with the chronic gastritiss gastric ulcer and cancer adjacent tissue.2The positive rate of Livin protein has no correlation with gender, age or lymph node metastasis, but with tissue differentiated degree (highly differentiated and poorly differentiated) and pathological types (adenocarcinoma and gastric signet ring cell carcinoma).3The gastric cancer patients weremostly syndromed of "Qi-blood deficiencysyndrome", and the positive rate of Livin protein in this syndrome was higher than other syndromes.4Livin was successfully transfected with the recombinant vector to MGC-803gastric cancer cell line and expression of Livin mRNA is detected by RT-PCR.5The cell proliferation ratio was significantly higher after Livin gene transfection.6Livin transfection group has a smaller proportion of cell in G0/G1state of cell cycle, a higher cell proliferation rate and a lower apoptosis rate than the other groups.ConelusionThe expression of Livin protein is closely related to gastric cancer and its pathological differentiation types. Livin gene transfection can promote gastric cancer MGC-803cell proliferation and inhibit apoptosis. Most of the gastric cancer patients are suffering from Qi-blood deficiency, Qi-blood deficiency might led to abnormal expression of Livin and induce the gastric cancer.
