Effect Of Baibankeli Ⅰ On PBMC Foxp3mRNA And TGF-β Protein Expression In Patients With Vitiligo

Objective:This study compared the level of PBMC Foxp3mRNA and TGF-β protein in serum between patients with vitiligo and normal control.We observed the level of PBMC Foxp3mRNA and TGF-β protein expression in patients with vitiligo and the changes before and after treatment, so as to analyze the pharmacological mechanisms of the immune with BaiBanKeLi I to reveal the molecular mechanisms of immune therapy of vitiligo.To provide clinical and theoretical foundation for the development and promotion of new drugs.Methods:The levels of PBMC Foxp3mRNA from patients with BaiBanKeLi I and the control group before and after treatment and healthy controls was detected by Real-time quantitative PCR analysis; The levels of TGF-β protein in serum from patients with BaiBanKeLi I and the control group before and after treatment and healthy controls was detected by ELISA analysis;and analyze the laboratory indicators with statistical methods.Result:1.The levels of PBMC Foxp3mRNA from patients with BaiBanKeLi I and the control group before and after treatment and healthy controls was detected by RT-PCR analysis.(1)RT-PCR test results show that Foxp3mRNA in vitiligo group was obviously lower than the normal control group, the difference was statistically significant.(P<0.05)(2)RT-PCR detection results showed that the treatment group the expression of Foxp3mRNA levels after3months is significantly higher than before treatment, the difference was statistically significant (P<0.05).(3) Control group3months after treatment the expression of Foxp3mRNA level was higher, the difference was statistically significant (P<0.05).(4)The changes of the expression of Foxp3mRNA between patients with BaiBanKeLi I and the control group before and after3months of treatment, the difference was statistically significant (P<0.05).2.The levels of TGF-β protein in serum from patients with BaiBanKeLi I and the control group before and after treatment and healthy controls was detected by ELISA analysis.(1) ELISA test results show that TGF-β in vitiligo group was obviously lower than the normal control group, the difference was statistically significant(P<0.05).(2)ELISA detection results showed that the treatment group the expression of TGF-β levels after3months is significantly higher than before treatment, the difference was statistically significant (P<0.05).(3)Control group3months after treatment the expression of TGF-β level was higher, the difference was statistically significant (P<0.05).(4)The changes of the expression of TGF-β between patients with BaiBanKeLi I and the control group before and after3months of treatment, the difference was statistically significant (P<0.05).Conclusion:1.The test results show that Foxp3mRNA and TGF-β in vitiligo group was obviously lower than the normal control group, the difference was statistically significant(P<0.05).We suggest that Treg cells have dysfunction in vitiligo.2.The changes of the expression of Foxp3mRNA and TGF-β between patients with BaiBanKeLi I and the control group before and after3months of treatment, the difference was statistically significant (P<0.05).BaiBanKeLi I can elevate the serum level of TGF-β and Foxp3transcription factor to normal or near normal levels in the patient with vitiligo by regulating the function of regulatory T cell,So BaiBanKeLi I can regulate the function of immume system.
