Objective:To amplify9Y-chromosomal Short Tandem Repeats (STR) locis in three multiplex systems by Silver Staining, which to investigate the genetic polymorphism in Korean and Han population from Yanbian area in China. Aim at offering data for Individual identification, Paternity identification, Human Genetics and other objects.Methods:Tested9Y-STR locis of200unrelated Korean and200unrelated Han individuals by PCR and analyzed by polyacrylamide gel electrophoresis with Silver staining. In this paper, DYS385a/b loci were analyzed by single-primer amplification technique, and the series of DYS450loci were analyzed by three multiplex systems. The Arlequin ver3.11and Excel were applied to calculate and analysis the genetics statistics of populations.Results:There were53,3,5,4,3,4,7,9,10alleles detected from Korean population in DYS385a/b, DYS450, DYS452, DYS453, DYS454, DYS455, DYS456, DYS458and DYS459a/b loci, and there were15,3,7,5,4,5,7,9,7alleles detected from Han population in DYS385a/b, DYS450, DYS452, DYS453, DYS454, DYS455, DYS456, DYS458and DYS459a/b loci; The Gene Diversity or Haplotype Diversity of9Y-STRs in Korean population were0.9648,0.2286,0.6607,0.1607,0.2097,0.0871,0.5074,0.7954,0.7001, and the Gene Diversity or Haplotype Diversity of9Y-STRs in Han population were0.9711,0.3355,0.6964,0.2510,0.2155,0.1692,0.5875,0.8163,0.5647; There were170haplotypes detected from Korean population, its Haplotype Diversity(HD) of accumulated9Y-STRs was0.9975, and there were188haplotypes detected from Han population, its Haplotype Diversity(HD) of accumulated9Y-STRs was0.9993; Moerover, compared two groups with differents ethnic in different areas, for obtaining P-value and Fst.Conclusion:DYS385a/b, DYS452, DYS456, DYS458and DYS459a/b loci were highly diverse Y-STRs loci and valuable in Individual identification and Paternity identification in Yanbian area, but other four locis were not; The data of9Y-STRs in two populations in Yanbian area can be provided in forensic Individual identification, Paternity identification, Human Genetics and other objects; The study on the comparation this two groups and differents ethnic in different areas confirmed that they had difference among different ethnic in different areas populations.
