Objectives:To investigate the expression of Hedgehog (Hh) signaling pathway related factors and cancer stem cell markers in cervical cancer Hela cells, and to study the ability of sphere formation with hypoxia.Methods:Hela cell was cultured with CoCl224h and48h, and than using quantitative RT-PCR and Western-Blot method to detect hypoxia inducible factor (HIF1a and HIF2a), Hh signaling pathway associated factor (smo, Gli-1) and tumor stem cell markers (oct-4, sox2, Nanog); sphere formation assay using checks Hela cells colony forming ability.Results:In cervical cancer Hela cells, the expression of HIF1a and HIF2a was significantly increased from hypoxia24h. Smo and Gli-1mRNA expression was increase from hypoxia48h; when hypoxia72h, Smo mRNA expression was5.4times of normoxia group (p <0.01, p<0.01), and Gli-1mRNA was123times of normoxia group (p<0.001, p<0.001). Cancer stem cells (CSCs) marker mRNA were increase from hypoxia24h in cervical cancer HeLa cells, and the level was significantly increased according the hypoxia extension times. At hypoxia72h, Oct4mRNA was78.8times of normoxia group, Sox2mRNA was787.4times of normoxia group, Nanog mRNA was689.8times of normoxia group (p<0.001, p<0.001, p<0.001). The ability of sphere formation in hypoxia group was significantly higher than in the normoxia group (p<0.01).Conclusion:Hedgehog signaling pathway was activated in Hela cells, and induced HeLa cell to cancer cell stemness with hypoxia.
