Objective: This study was designed to investigate the mechanism underling effects ofBushen Zengmo prescription on tissue morphology and expressions of ERs and PRs inendometrium of mice superovulated by GnRHa, in order to providing scientific basisfor the role of this prescription in improving the receptivity of endometrium.Methods: Experimental animals were randomly divided into5groups with each10:blank control group, model control group, positive control group, low Bushen Zengmogroup, and high Bushen Zengmo group. Through intragastric administration, thecontrol group was treated with0.9%saline by0.02ml/g·d, model control group withGnRHa for establishing superovulation model, positive control group with0.02ml/g·daspirin besides GnRHa, low Bushen Zengmo group with a low dose of Chinesemedicine besides GnRHa (10folds than human usage), and high Bushen Zengmogroup with a high dose of Chinese medicine besides GnRHa (20folds than humanusage). After11days successive administration, the animal model was established.48h later, all mice were injected with0.2ml HCG. Smears of vaginal exfoliative cellsreveal the statement of ovulation. After cervical dislocation, the uterus was removedand its upper and middle parts were fixed by4%paraformaldehyde. The expression ofERs and PRs were stained by SP immunohistochemical method, and the morphologicchanges in endometrium were observed by HE preparation.Results: Results showed that there is no statistical difference between the blank andlow Bushen Zengmo group(P>0.05), while compared with the rest group, theexpression of both ERs and PRs in the control group differed significantly (P<0.01).In model control group, the expressions of ERs and PRs presented a significantincreased (P<0.01) with the comparison to the rest groups. Statistical significanceoccurred when positive control compared with the rest groups expect for low Bushen Zengmo group. ERs and PRs contents were enhanced in high Bushen Zengmo groupwith P<0.01, when compared with other groups. These findings indicated that there isa direct relationship between Bushen Zengmo prescription and the increased ERs andPRs expressions in endometrium of mice superovulated by GnRHa, and also revealedthat Bushen Zengmo prescription functions in improving the receptivity ofendometrium.By the use of light microscope, in control group abundant and uniform glands,big glandular cavity, loosened and dropsical mesenchyme were observed in mouseendometrium. Nevertheless, less endometrial glands, smaller glandular cavity, andmore tight mesenchyme occurred in model group. Mice administrated withBushengzengmofang showed alleviative pathological symptom, suggesting that thecompound prescription can promote the synchronization of glands and mesenchymewhich makes for the blastocyst planting, especially high dose.Conclusion: Busheng Zengmo prescription could elevate the expression of ERs andPRs endometrium of mice with controlled superovulation, optimize the morphologyand function of endometrium, and enhance the receptivity of endometrium in micewith superovulated by GnRHa, the effect is better than that of aspirin,safe andreliable.
