| Objective: to observe the kidney detoxification t2dm party of Wistar rats withexperimental autoimmune encephalomyelitis (experimental autoimmune encephalomyelitis,EAE) of clinical manifestation, pathological changes and the onset process interferon (CNS)-the effect of gamma, its effect on prevention and control of MS is discussed and themechanism of action, for the development of new treatments for multiple sclerosis way toprovide meaningful basic experiment basis.Methods:(1)75in healthy female Wistar rats, were randomly divided into: blankgroup, model group, Chinese medicine treatment group, Chinese medicine treatment groupand the dose divided into low dose treatment group, middle dose group, high dose treatmentgroups, each group of15only. Blank group will not do any processing; Model group beganafter giving saline every day0.2ml building to fill the stomach; Immune day started totraditional Chinese medicine group, high-dose group of60g per kilogram of body weightper day to give Chinese medicine content2ml lavage, dose group per kilogram of bodyweight every day30g to1ml lavage content, low dose group of15per kilogram of bodyweight per day to give0.5ml lavage, content above lavage method is to fill the stomach toterminate experiment.(2) the EAE model: the guinea pig spinal cord homogenate and complete the type ofadjuvant (CFA) made of a mixture as antigen, intradermal injection of pertussis vaccine atthe same time, in addition to the blank group. Daily observation of the clinicalmanifestations of the groups of animals after building, measuring weight change, and givethe score of neurological function.(3) samples:14days and21days after immunization, respectively, using the eyesocket after venous plexus in pursuance of law to take venous blood, heart perfusion in brainand spinal cord after put in4%paraformaldehyde fixed, paraffin embedding, sectioning.(4) wood grain-eosin (HE) staining to observe the brain, spinal cord pathologicalchanges; Within the ELISA method for determination of serum interferon-gamma content.Results:(1) the kidney detoxification t2dm party the incidence of high, middle dosegroup of rats compared with EAE group significantly reduced (P <0.01), kidneydetoxification t2dm with high, middle and low group the average incubation period of ratssignificantly longer in the EAE group (P <0.01); Kidney detoxification t2dm party high, middle dose group of rats pathogenesis peak, remission of nerve function score significantlylower than the EAE group; Kidney detoxification agent, high dose group of t2dm party peakincidence in rats, weight of remission was obviously higher than that of EAE group, lowdose group.(2) the onset of kidney detoxification t2dm with high, middle, low dose group of ratCNS inflammatory lesions in number compared with EAE group significantly reduced (P <0.01), the remission kidney detoxification t2dm rat CNS side of high, middle dose group ofseveral inflammatory lesions compared with EAE group significantly reduced (P <0.01),low dose group compared with EAE group no significant difference (P>0.05).(3) the kidney detoxification t2dm with high, middle, low dose group of incidencepeak serum interferon-gamma levels decreased significantly in the EAE group (P <0.01);Kidney detoxification t2dm party high, middle dose group of disease remission-seruminterferon gamma levels decreased significantly in the EAE group (P <0.01), low dosegroup was significantly higher than that of high dose group (P <0.05), high, middle dosegroup had no significant difference (P>0.05).Conclusion:(1) the kidney detoxification t2dm party preventive treatment canobviously reduce the incidence of acute EAE rats, relieve the clinical symptoms, weight loss,shorten the course of the disease.(2) the kidney detoxification t2dm party preventive treatment can reduce incidencenerve function damage degree, reduce EAE rat CNS inflammation.(3) the kidney detoxification t2dm party part of the action mechanisms of preventionand treatment of MS could be cut in the serum of rats by the expression of interferon-gamma, suppressing inflammation and immune reaction.(4) the kidney detoxification t2dm with high, middle dose effect is not obviousdifference. |
