Objective:Research on the effect of genuine se-men ziziphi spinosae to theanxiety behavior in the Yin-deficiency rats and the body distribution ofmajior components.Methods:1The rats in the Yin-deficiency model group,diazepam group andsemen ziziphi spinosae group were given the suspension of thyroid tablet160mg·kg-1continue13days to prepare the yin deficiency model,and werebegan to give medicine on the eighth day afternoon of building,amongthem,the rats in diazepam group were given the suspension of diazepamtablets2.0mg·kg-1,and the rats in semen ziziphi spinosae group were giventhe water decoction of semen ziziphi spinosae10g·kg-1. Evolution thefunction of genuine se-men ziziphi spinosae in the yin deficiency rats byobserving the behavior changes of rats,the food-intake and water intake of24hours in rats,the weight difference before and after the experiment andthe organ coefficient and the measurements of T3,T4,TSH. Prepare the yindeficiency anxiety composite model by combining with elevated cross mazedevice and light box device, and evolution the anti-anxiety effects ofgenuine se-men ziziphi spinosae by contrasting the open arms lengthpercentage (OT) and the number percentagy into the open arms (OE%) ofrats in elevated cross maze device,and contrasting the back and forthnumber (TT) of rats in light box device.2Select the rats in the group of semen ziziphi spinosae, first ofall,pick the eyeball and take the blood after the rats were given medicine 10,20,30,40,60,90,120,240minutes,then execute the rats and collect theheart,liver,spleen,lung,kidney,stomach,brain,large intestine and smallintestine and so on, afterwards, determine the concentration ofspinosin,jujuboside A and jujuboside B in different tissues byHPLC-PDA-ELSD after the tissues were homogenated, finally, get therelative pharmacokinetic parameters by using DAS2.0software to deal withthe detected data previously mentioned.Results:1Compared with the group of yin deficiency model, semen ziziphispinosae can significantly reduce the abnormal increased food-intake(p<0.01) and water intake (p<0.01) of24hours in rats,the kidneycoefficient (p<0.01),adrenal coefficient (p<0.01),the value of T3(p<0.01)and T4(p<0.05); and significantly increase the weight difference (p<0.01)of rats before and after the experiment,the value of TSH (p<0.01),thenumber percentagy into the open arms (p<0.01),the open arms lengthpercentage (p<0.01) and the back and forth number (p<0.01) of rats in lightbox device, show that semen ziziphi spinosae have obvious resistance to yindeficiency and anti-anxiety effect.2The body distribution of spinosin、 jujuboside A and jujuboside B inrats: after the rats were given water decoction of semen ziziphi spinosae,spinosin and jujuboside A were widely distributed inblood,heart,liver,spleen,lung,kidney,stomach,brain,large intestine and smallintestine,and these two components can be detected after the rats were givenmedicine10miniutes and sustainable to240miniutes; but jujuboside B onlydistributed in blood,stomach,large intestine and small intestine.The order of average concentration in the organization of spinosin issmallintestine>stomach>liver>brain>largeintestine>spleen>lung>heart>kidney>serum, and the order of AUC0-tin the organization of spinosin is smallintestine>stomach>liver>largeintestine>spleen>brain>heart>kidney>lung>serum; The order of average concentration in the organization of jujubosideAislung>largeintestine>heart>speen>liver>kidney>smallintestine>stomach>brain>serum, and the order of AUC0-tin the organization of jujuboside Ais lung>spleen>liver>heart>large intestine>brain>stomach>kidney>smallintestine>serum; The order of average concentration in the organization of jujuboside B is large intestine>stomach>small intestine>serum, and theorder of AUC0-tin the organization of jujuboside B is largeintestine>serum>small intestine>stomach.Conclusion:Water decoction of semen ziziphi spinosae can effectively improvethe symptoms of yin deficiency,and can well resist the anxiety behavior ofthe yin deficiency rats, which show that water decoction of semen ziziphispinosae have a good effect on anxiety. The tissue distribution of threetypical composition of genuine se-men ziziphi spinosae in the rats whitchwere yin deficiency and anxious is close to the channel tropism of genuineSe-men Ziziphi Spinosae.
