1Background and ObjectiveEpidemiological studies showed that about the two thirds of hypertensivepatients have suffered from type2diabetes mellitus (T2DM). Reason for theincreased incidence is a research hotspot at present. A series of studies have shownthat endogenous glucose production increase in people with T2DM is mainly due togluconeogenesis level increased. RAS system is activated, sympathetic hyperactivityoccurs when hypertension exist, which will certainly affect glucose metabolism, buthow much will it affect the body’s level of gluconeogenesis need further research.In this study, spontaneously hypertensive rats (SHR) and homologous Wistar ratswere used as the research object. By comparing the levels of G-6-Pase and PEPCKmRNA expression in SHR and Wistar rats, we may understand the impact ofhypertension on gluconeogenesis.2Methods2.1GroupingIn this experiment, there are two species of rats,16weeks-aged SPF male SHRrats (20) and Wistar rats (20). All Rats have adaptive feeding for one week before thetest. And then the SHR rats were randomly divided into two groups,S1(10) and S2group (10); Wistar rats were also randomly assigned to W1group (10) and W2group(10). 2.2Feeding methodThe rats in W1and S1group were given normal diet, and the rats in W2and S2group were fed with high-fat&high-sugar (HFHS) diet for a total of12weeks. Keeping the environment temperature of22-27℃, humidity was maintained at50%±10%, day and night time control (1:1) and free eating and drinking.2.3Detection index2.3.1The fasting plasma glucose,insulin,TG,TC and LDL-C have been examined in the end of the experiment;2.3.2The of islet function and insulin resistance degree were evaluated by glucosetolerance test, insulin release test and insulin resistance index (HOMA-IR);2.3.3The Glucose-6-phosphatase (G-6-Pase) and phosphoenolpyruvate carboxylasekinase (PEPCK) gene expression level was detected by RT-PCR.3Results3.1Blood chemistryThe level of TC, TG, LDL-C in the W2and S2group increased significantlycompare to their respective control groups(W1and S1), the difference was significant(P <0.05); The above index in S1group is higher than that in W1group, but thedifference were not significant (P>0.05); The level of TC and LDL-C in S2group ishigher than that in the W2group, the difference was significant (P <0.05).3.2Insulin resistance indexThe HOMA-IR in the W2group is higher than that in the W1group(P <0.05),and the HOMA-IR in the S2group is higher than that in the S1group(P <0.05), thedifference was significant. Compared with W2group, the HOMA-IR of S2groupincreased obviously, but there was significant difference between two groups (P <0.05).3.3The expression levels of PEPCK and G-6-Pase mRNA in Liver and kidneyAfter12weeks of HFHS diet, the expression level of PEPCK and G-6-Pase mRNA in the W2group increased more obviously than that in W1group, there wassignificant difference between two groups (P <0.05), and the levels have been raised2times in Liver and3times in the kidney. Compared with Wistar rats, the expressionlevels in SHR rats were also significantly increased (P<0.05); 4Conclusions4.1Long term HFHS diet can lead to PEPCK and G-6-Pase mRNA expression levelsincrease;4.2The expression levels of PEPCK and G-6-Pase mRNA increased in SHR rats bothin normal diet and HFHS diet were more obvious than that in the Wistar rats;4.3The increased degree of PEPCK and G-6-Pase mRNA expression in the kidneywas higher than that of liver in HFHS diet feeding rats.
