Background and PurposeEndometrial polyps are vegetations protruding from the surface of the endometrium,as one of the common benign gynecological diseases. EP can cause abnormal uterinebleeding, in women of reproductive age, mainly excessive menstrual intermenstrualbleeding or infertility, in postmenopausal women is shown as irregular bleeding. EP iscomposed of glandular and stromal components, glandular part often show simplehyperplasia or cystic glandular hyperplasia, surface coating of glandular epithelium,interstitial part are composed by a small dense fibrous connective tissue,inflammatory cellinfiltration and blood vessels with thicker wall. In recent years, due to the wide applicationof improving the quality of people’s lives and hysteroscopy, the incidence and detection rateof EP increased year by year, it impacts the modern women’s physical and mental health. Atpresent, the pathogenesis of EP is not yet completely clear, one important pathologicalfeatures of the stroma containing thick-walled blood vessels, therefore the prediction maybe associated with some blood vessel related production factors.Mast cells derived from bone marrow hematopoietic stem cells, is a kind of commonimmune response cells, known as the "multifunctional cytokine source". Recent studieshave found that many angiogenesis related diseases rely on mast cells, it can stimulate the proliferation and angiogenesis in endothelial cells,this function is mainly done by MastCell Tryptase(MCT). As a specific marker of MC, it is a kind of active neutral proteasewhich presynthesized in MC. MCT-β2is a major protease in degranulation of MC, theexpression of MC and MCT in EP has not been reported at home currently.Transforming growth factor-β1is an important factor in promoting and regulatingangiogenesis, TGF-β1can directly stimulate endothelial cell proliferation, chemotaxis andmigration, and also be inducted by VEGF and other factors to promote the formation ofnew blood vessels. It plays an important role in promoting cell proliferation, extracellularmatrix deposition, angiogenesis, inflammatory cell infiltration and tissue fibrosis process.So far, Endostatin is the strongest endogenous angiogenesis inhibitor, which candirectly inhibit vascular endothelial cell proliferation, the growth cycle arrest, apoptosis,butalso inhibit endothelial cell proliferation and migration by several vascular growth factorssuch as VEGF,bFGF and also stimulate MMP, degrade of the extracellular matrix, causemigration of endothelial cells. ES have no inhibition effect on non-endothelial cells.Therefore, this article discusses the characteristic expression of MCT-β2, TGF-β1andES in endometrial polyps, correlation analysis of these factors, it speculated that therelationship between these factors and the formation of new blood vessels, for theunderstanding of the pathogenesis of EP and the postoperative recurrence, antiangiogenesis therapy to provide a theoretical basis.Material and Methods1.GroupingPolyp Group: Select45cases of patients with endometrial polyps tissue underwenthysteroscopic resection of endometrial polyps during the period of April to October2013inour hospital.Normal endometrium Group: Select17cases of patients with tubal infertility byhysteroscopy, with the informed consent, take a small amount of normal endometrial tissueunder the microscope, the pathological results for proliferative phase endometrium.2.MethodsThe Immunohistochemical SP technique was used to detect the protein expression ofMCT-β2, TGF-β1and ES in polyp tissues and normal endometrial tissues.3.Statistical analysis All of experimental data input SPSS17.0package deal. The positive rate of MCT-β2,TGF-β1and ES expression in polyps and normal endometrium tissues were compared byusing χ2test, correlation analysis using correlation analysis of2×2paired data. Test level is0.05(bilateral).Results1. The positive expression of MCT-β2in polyps group was higher than that in normalendometrium group, there was significant difference (P<0.05);2. The positive expression of TGF-β1in polyps group was significantly higher thanthat in normal endometrium group, there was statistically significant difference (P<0.05);3. The positive expression of ES in polyps group was significantly higher than that innormal endometrium group, the difference was significant (P<0.05);4. The expression of MCT-β2and TGF-β1proteins in endometrial polyps is positivelycorrelated (P<0.05, r=0.327);5. The expression of MCT-β2and ES proteins in endometrial polyp tissues ispositively correlated (P<0.05, r=0.485);6. TGF-β1and ES protein has no obvious correlation in endometrial polyp tissues(P>0.05).Conclusions1. As a specific marker for MC, MCT-β2is higher expression in the endometrialpolyps, suggesting that MC is involved in the formation of endometrial polyp of thick wallvessels, plays an important role in the proliferation process of polyps.2. MCT-β2and TGF-β1in endometrial polyps show high expression. They arepositively correlated, both of them jointly promote the occurrence and development ofendometrial polyps.3. ES is highly expressed in endometrial polyps, it is positively correlated with theexpression of MCT-β2, suggesting that it may be related to angiogenesis regulation ofcompensatory effects in vivo.
