Background and purposeNeurogenic bladder (Neurogenic Bladder, NB) is a common urologicaldiseases. Spinal reflex center, sympathetic, parasympathetic nerve activityparticipation and control urination, bladder voiding any related neuropathy or(and)damage caused by bladder or (and) urethral dysfunction, often accompanied byurinary bladder function coordination disorders, referred to as neurogenic bladder[1].Neurogenic bladder urinary system diseases treatment of the most complex anddifficult clinical conditions, either the pathogenesis or clinical treatment, there arestill many issues that we need to solve. Therefore, it is necessary to further study. However, in view of medical ethics and other reasons, many studies conducted inpatients who can not directly establish an animal model studies on greatsignificance.NB most established animal model of spinal cord injury or spinal cordtransection choice, Mainly caused by neurogenic changes (mainly the followingurinary tract symptoms), Spinal cord transection modeling methods and modelingmethods Allen’s against spinal cord injury compared to the disadvantage ofcomplicated surgical operation, large amount of bleeding, the operation can not be repeated. According to reports, Allen’s heavy fall to combat injury in animal modelscan simulate the impact of the characteristics of the human spinal cord injury[2]. Inthis study, this simple method of spinal cord injury model, intended to establish ananimal model of neurogenic bladder and urodynamic evaluation to explore the safetyand stability of a strong, fast and simple, reproducible method of making high nerveinjury model. clinical studies detrusor contraction pathophysiological neurogenicbladder to provide a reference.Research Subjects and MethodsFemale Wistar rats of clean grade22(First Affiliated Hospital of ZhengzhouUniversity, animal breeding centers, aged4-6weeks, weight (200±20) g), set oneweek before surgery feeding cage so familiar with the environment. BBB motorfunction assessment conducted two groups of rats,Observer (non-experimental groupmembers) will be rats placed on the test bench, rats began to adapt to until theindependent observation for10minutes. Grouping: NB group (n=14). Normalcontrol group (sham group, n=8). NB groups using modified Allen’s method to T12as the center back median longitudinal incision to expose the T11~T13spinousAAprocess and lamina, spinous process and the bite of the vertebral segmentsremoved, exposing the corresponding spinal segment as the damage zone, the first inthe hard place the top of a rectangular plastic film spacer (0.4mm×0.3mm), thenusing quantitative units Allen’s WD method using5g weight at the height of10cm(injury of [5]:5g×10cm=50gcm) Department, through the catheter free fall, weight spinal cord stem end down quantitative fight to establish spinal cord injurymodel. NB and normal control group BBB assessment of motor function.Surgicaloperations as normal control group, but only after the bite except spinous layer woundclosure.Video-urodynamic assessment was performed at5-6days post operation.Using SPSS17.0statistical package for statistical analysis.Measurement data±sdescription of the two samples were compared using independent sample t test.Inspection standards set0.05, P <0.05was considered statistically significanceResultssurgery After the NB group and the control group were Wistar rats torsoappears soft, insensitive reaction, do not eat. NB group in rats after surgery hindlimb drag state, quiet and shortness of breath.2-6days after surgery NB group (n=14)always showed hind limbs and tail could not. The control group showed nosignificant abnormality., without neurological damage. NB spinal cord injury in ratswere urinary retention and varying degrees of difficulty urinating.NB group and thesham group BBB score, there are significant differences were statistically significant(P <0.05). NB rats after5-6d display images urodynamic bladder volume increases, atypical "football" shape, the control group bladder radiological images show nosignificant increase in bladder volume. Detrusor showed areflexia, maximum bladdercapacity(MBC) and leak detrusor pressur(LDP) and bladder compliance (BC)increased significantly in NB group, in NB and control group the MBC was3.93±2.03ml vs.0.081±0.247ml,BC was0.093±0.54ml vs.0.022±0.01ml, LDP was45.38±12.52ml vs.26±3.85ml respectively,(P <0.001).ConclusionThe modified method of Allen’s is useful to establish NB.
