Objective: To explore the effect of total flavones from Litchi chinensis Sonn onliver function and the expression of NF-кB and type Ⅲ procollagen with fibroticrats induced by common bile duct ligation. Methods:60SD rats were randomlydivided into six groups (n=10each): bile duct ligation group (BDL group), TFLlow dose group, TFL medium dose group and TFL high dose group, silibiningroup (SIL group), sham-operated group (SO group) as a negative control. Thehepatic fibrosis model was established by common bile ductligation.Simultaneously, rats in TFL low, medium, high and SIL groups wereperfused with TFL or SIL respectively for four weeks. The BDL group and SOgroup were perfused with saline for four weeks. All animals were sacrificed at theend of the fourth week after operation, serum level of superoxide dismutase (SOD),alanine aminotransferase (ALT), aspartate aminotransferase (AST), direct bilirubin(DBIL) and total bilirubin (TBIL) was measured. Pathological changes in liverwere evaluated by hematoxylin-eosin (HE) and Masson staining. Expression ofNF-кB and PCⅢ was detected by immunohistochemistry and Western blotting.Results: Compared with BDL group(147.35±86.27)IU/L, there was obviousdifference between TFL medium and high dose groups(92.93±47.27)IU/L、(77.16±29.45)IU/L inALT level (t=2.24、3.06,P<0.05).The serumAST level inTFL high dose group(218.34±139.77)IU/L was significantly lower than that in the BDL group(362.73±106.61)IU/L,(t=3.23,P<0.05).Compared with the SILgroup(137.01±38.86)μmol/L, there was obvious difference between TFLmedium and high dose group(81.48±47.50)μmol/L、(50.06±53.99)μmol/L inDBIL level(t=3.33、4.62,P<0.05).The serum TBIL level in TFL high dose group(63.33±69.99)μmol/L was significantly lower than that in the BDL group(132.51±52.24)μmol/L and in SIL group (151.82±60.40)μmol/L,(t=2.57、3.62,P<0.05).Compared with SO group(215.45±14.94)U/mg, there was nosignificant difference between TFL medium and high dose group (207.58±17.91)U/mg,(197.74±13.53)U/mg in serum SOD level(P>0.05).Comparedwith BDL group(185.14±26.9)U/mg, there was no significant differencebetween SIL group and TFL low group(195.39±23.8)U/mg,(194.15±27.77)U/mg in serum SOD level(P>0.05).There was significant difference among sixgroups on protein expression of NF-кB in liver tissue (F=1168,P<0.05).Theexpression of NF-кB and PCⅢ protein in liver tissue decreased gradually inSIL group, TFL medium dose group and TFL high dose group, Compared withBDL group, there was significant difference among groups described above(F=450.89,P<0.05).Conclusions: TFL with some dose has a powerful antioxidant,improves liver function in rats with cholestatic liver fibrosis, inhibits the progressof liver fibrosis; Its mechanism may be related to inhibiting the expression of NF-кB and PCⅢ protein in liver tissue.
