Objective: Puppies of extracorporeal circulation model is established,under the open-heart surgery, continuous perifision and continuousperfusion of immature myocardial protective effect and its possiblemechanism.Methods: Will only younger than8weeks with20species, close tothe weight of healthy puppies were randomly divided into reality accordingto the group with a pair of, through the establishment of animal model ofcardiac arrest atfer cardiopulmonary bypass surgery puppy, comparing thecontrol group (n=10, continuous extracorporeal circulation2hours) andexperimental group (n=10, extracorporeal circulation for1hour, andthen continue atfer1hour) puppies myocardial damage index (enzyme linked immunosorbent test for determinsation of serum cTn-I. ANPcontent), cardiac ultrastructure changes (electron microscope) andmyocardial cell apoptosis,(the ends of DNA nucleotides transferasemediated Nick end labeling method, method of TUNNEL), myocardialcells of caspase3and survivin protein expression level difference.Measurement data with mean土standard deviation, said the comparison ofmeasurement data using T test between the two groups; Count datacomparison with rank and inspection; Multiple indicators related toinspection of the contact, with P <0.05said the difference was statisticallysignificant. The above statistics processing using SPSS13.0statisticssotfwareResults:During the CPB, experimental group and control group inserum cTn-1, ANP content were higher, but the higher the experimentalgroup were lower than the control group; Experimental group and controlgroup in the cardiac ultrastructure were disruptive change, but in theexperimental group was slight damage; Experimental group and controlgroup in the myocardial cell apoptosis occurred, but the experimentalmyocardial apoptosis number less than the control group; Caspase3proteinexpression in the experimental group and control group were higher, butthe control group increased more significantly; Survivin protein expression in the experimental group and control group were higher, but theexperimental group increased more signiifcantly; In the same group, themyocardial cells of caspase3and negatively correlated with survivin proteinexpression.Conclusions: Puppies open-heart surgery after irrigation can improvemicrocirculation under CPB puppies, oxygen supply, improve the puppyreduce MIRI; Make high survivin protein expression in myocardial tissue,relatively lower caspase3protein expression of myocardial cells (caspase3,and negatively correlated to the expression of survivin protein), caneffectively reduce the myocardial cell apoptosis, the immature myocardialprotection.
