ObjectiveThe aim of the study is to repair SD rat sciatic nerve crush injury with Neurotubenerve guidance conduit, and observe its sciatic nerve functional recovery to investigate theNeurotube nerve guidance conduit whether to repair the peripheral nerve crush injury.Methods60healthy adult SD rats, male, weight200-250g, after modeling were randomlydivided into simple sciatic nerve injury group (n=20), nerve suture group (n=20) and theNeurotube nerve guidance conduit repairing group (n=20).After1,2,4,6weeks of theoperation,5rats in each group were taken randomly to measure sciatic nerve functionindex and local nerve growth factor (NGF) density,and detect each regeneration of nerveaction potential latency, amplitude and conduction velocity, document electrophysiologyresults.ResultsSciatic nerve function index, no significant difference exists between the three groups(P>0.05) after one week of the operation,that demenstrate the injury of sciatic nerve nodifference.In1,2,4weeks, the nerve suture group is better than the Neurotube nerveguidance conduit repairing group, the results were statistically significant (P<0.05),however, there was no significant difference between the two groups at the6weeks(P>0.05). The local density of NGF, the Neurotube nerve guidance conduit repairinggroup were higher than the simple injury group and the nerve suture group, the results were statistically significant (P<0.05). After6weeks of electrophysiological examination,the Neurotube nerve guidance conduit repairing group was significantly better than thesimple injury group, the difference was statistically significant (P<0.001); but theNeurotube nerve guidance conduit repairing group and nerve repair groups was notstatistically significant (P>0.05).Toluidine blue-stained showed the simple injury group ofnerve fibers of varying sizes, irregular shape of myelin,axon swelling, Schwann cellproliferation; the nerve suture group showed more regenerating nerve fiber,myelin small,oval or triangular, axon fine, residual degeneration of nerve fibers in the nerve fiberregeneration; the Neurotube nerve guidance conduit repairing group of nerve fibers had thesame size, and myelin, axon located in the central.ConclusionsNeurotube nerve guidance conduit can promote the repair of SD rats sciatic nerveinjury.
