Objective To observe The expression of p-JAK2and p-STAT3and cell apoptosis of the brain damage following cerebral ischemia/reperfusion in rats that were injected recombinant huaman granulocyte colony-stimulating factor (rhG-CSF);And explore the neuroprotective mechanisms of rhG-CSF.Methods Thirty-six male Sprague-Dawley rats were randomly divided into the sham-operated group,the middle cerebral artery occlusion/reperfusion group (MCAO/R) and the rhG-CSF-treatment group, and each group had12rats. The temporary middle cerebral artery occlusion reperfusion model was made by the suture method. In rhG-CSF-treatment group, rats were injected a single dose of50μg/kg rhG-CSF subcutaneously at two hours and24h after the reperfusion. The sham-operated group and MCAO/R group received the same volume of saline. The neurological function was scored with Longa5-point scale. The expressions of p-JAK2、 p-STAT3were detected by immunohis toch emistry. The apoptosis cells were shown by TUNEL method. The infarction volume was revealed by TTC stain.Result the infarction was observed in the cerebral ischemia-reperfusion group and the rhG-CSF-treatment group. The infarction volume (16.27±1.44%) and neurological outcome scores of the rhG-CSF-treatment group were lower than the ischemia-reperfusion group.the mean gray values of the expressions of p-JAK2and P-STAT3in the rhG-CSF-treatment group(36.47±6.38and35.±5.52,) were significantly increasde compared with ischemia-reperfusion group (25.62±6.25and24.52±5.08,P<0.01,respectively).Compared with the cerebral ischemia-reperfusion group(39.52±3.74%), the percentage of apoptotic cells decreased significantly in the G-CSF-treatment group (27.84±2.69%,P<0.01respectively).no infarction was observe and apoptotic cells were hardly to be found In the sham-operated group, the mean gray values of the expressions of p-JAK2and p-STAT3in the sham-operated group (p-JAK2(41.59±6.43),p-STAT3(43.36±5.68)) were higher than ischemia-reperfusion group (p-JAK2(25.62±6.25),p-STAT3(24.52±5.08),P<0.01)Conclusion rhG-CSF could protect brain from ischemia/reperfusion injury,and the neuroprotective mechanism of rhG-CSF may inhibit inflammatory cytokines, mediate the JAK2/STAT3signal pathway to reduce nerve caused by cerebral ischemia reperfusion injury and nerve protective effect.
