OBJECTIVE To study the protection of Edaravone on the oxidatie stress injury caused by the cerebral ischemia-reperfusion in rats and its effect on apoptosis activity of hippocampus cells.METHODS Wistar rats were randomly divided into three groups according to their weight:false operation group, ischemia-reperfusion group, and Edaravone group; acute cerebral ischemia model in rats was preparated using middle cerebral artery suture occlusion. After acute global cerebral ischemia model law prepared successfully by MCAO, group edaravone within30min after intraperitoneal injection of edaravone3mg/kgwt,30min after subcutaneous injection of the same dose given once, To maintain a stable blood concentration of drug treatment, Sham group and I/R group given the same method with normal saline. Give the rats once daily dosing, Repeat7d; Determination of infarct area (TTC) staining. Using immunohistochemical techniques to detect the amount of brain hippocampus tissue Caspase-3, Bcl-2, Bax expression.RESULTS1、Compared With cerebral ischemia-reperfusion group (I/R group), Edaravone (E group) can significantly improve brain tissue SOD activity (P<0.05). Group E:15.86±2.41vs group I/R;12.29±3.41, P<0.052、Compared With cerebral ischemia-reperfusion group (I/R group), Edaravone (E group) can significantly reduce brain tissue LDH activity (P<0.05). Group E:136.52±33.76vs group I/R:196.03±43.20, P<0.053、Compared With cerebral ischemia-reperfusion group (I/R group), Edaravone (E group) can significantly reduce brain tissue MDA activity (P<0.05). Group E:30.50±3.04vs group I/R:36.41±5.43, P<0.054、Compared With cerebral ischemia-reperfusion group (I/R group),Edaravone (E group) can reduce infarct volume (P<0.05), Group E:48.89±7.63vs group I/R:72.23±6.53, P<0.05。5、Edaravone for Bcl-2and Bax positive cells affect positive cells. Bax:group E:10.50±3.21vs group I/R:.16.41±3.32, P<005, Edaravone can reduce the number of Bax positive cells Bcl-2; Group E:.24.37±3.74vs group I/R:19.03±4.10, P<005, Edaravone can increase the number of Bcl-2positive cells. Showed that Bcl-2/Bax expression increased proportion (P<0.05).6、Compared With cerebral ischemia-reperfusion group (I/R group), Edaravone (E group) can significantly reduce Caspase-3positive cells (P<0.05). Group E:15.81±2.35vs group I/R:.22.29±3.31, P<005CONCLUSION Edaravone could protect the oxidatie stress injury caused by the cerebral ischemia-reperfusion in rats, and it could decrease apoptosis by raising the expression of Bcl-2and inhibiting the expression of Bax at the same time, inhibiting the expression of Caspase-3, so that it could protect brain tissue.
