Object:To investigate the mechanism of p53gene expression on Meso-Dihydroguaiaretic Acid inhibiting gastric cancer cell proliferation.Method:The gastric cancer cell MGC-803’s morphology changes were observed by phase-contrast microscope. MGC-803cells were treated with Meso-Dihydroguaiaretic Acid in vitro, and the cell viability was detected by using MTT assay. RT-PCR and western blot were used to detect the expression of p53in MGC-803cells treated with lignans. Meanwhile the cells untreated were detected as control.Results: By the microscope observation, gastric carcinoma cells of the control group were more vigorous and prolongation. While the MG-C803cells treated Meso-Dihydroguaiaretic Acid were depressed in the same period of time. MTT assay showed that the survival rate of the experiment group was significantly lower compared with the control group (P<0.01). The mRNA and protein expression level of p53dramatically downregulated in MGC-803cells treated with experiment compared with the control untreated cells. These results indicated thatp53played an important role on the inhibitory effect of Meso-Dihydroguaiaretic Acid on MGC-803cells by RT-PCR and western blotting assay (P<0.05).Conclusions:Meso-Dihydroguaiaretic Acid could effectively suppress the MG-C803cells in vitro via down-regulating the expression of p53by inhibiting the cell proliferation.
