Objective:To study the effect of abnormal expression of GPC5on the biological behavior of prostate cancer cells. And make a preliminary study into its molecular mechanis.Methods:1.Structre GPC5abnormal expression plasmi,transfect prostate cancer cell PC3and LNCap. Compared with the control group to observe and analyze the effects of abnormal expression of GPC5on the proliferation and invasion.2.By enhancing the expression of c-Myc in prostate cancer cell PC3,to observe the overexpression of c-Myc on the expression of GPC5in prostate cancer cell.3.Using Co-IP and mass-spectrometric technique,to find the growth factor/receptor ligand members in the prostate cancer cells.Results:1.Overexpression of GPC5significantly inhibites the proliferation, invasion of prostate cancer cell PC3.Low expression of GPC5significantly promotes the proliferation, invasion of prostate cancer cell LNCap.2. Overexpression of c-Myc significantly inhibits the expression of GPC5in prostate cancer cell PC3.3. Immune protein coprecipitation and plain analysis shows in prostate cancer cells is PC3, liver derived growth factor (HDGF) combined with GPC5and play a role.Conclusion:1. The abnormal expression of GPC5can influence the cell biological behavior of prostate cancer cell PC3and LNCap, confirmed that GPC5plays an important role in the occurrence and development of prostate cancer, and may become a potential target for the treatment of prostate cancer. 2. Overexpression of c-Myc inhibits the expression of GPC5in prostate cancer, then influence the development and progression of prostate cancer.3. GPC5on the prostate cancer cell membrane surface may be a co receptor of exogenous HDGF and mediates its enter cells and play a role.
