Objective:To establish a convenient and stable in vitro screening method ofhepatoprotective effect, then screen the hepatoprotective effect of the activecomponents of thermal transformation products from swertiamarin by using themethod. To clarify the material basis of the hepatoprotective effect of thermaltransformation products from swertiamarin, and reveal the antihepatitis relationshipbetween thermal transformation products from swertiamarin and Swertia.Methods:(1)Use acetaminophen (AP) reproduce L-02cell damage model, and choose theappropriate dosage and the action time of the positive control drug,N-acetyl-L-cysteine (NAC). Then judge the success of the damage model by detectingthe enzyme lactate dehydrogenase (LDH).(2)Screen the hepatoprotective effect of the active components of thermaltransformation products from swertiamarin by using AP-induced L-02damage modeland detecting the survival rate (MTT assay); measure the LDH activities in themedium to screening futher.Results:(1) AP-induced L-02cell damage model was successfully built. The IC50valueat48h was3.97mmol/L. Compared with model group the1.0mg/ml NACsignificantly improved the cell survival rate by about15%,and decreased the LDHactivity in the medium of model cells. (2)The results of the cell survical rate were detected by MTT assay, show thatdose within0.8-100μg/ml compared with model group, components5#、7#、8#、10#、12#、16#、20#、22#can improve the model cell survival rate; compared with modelgroup, NAC,8#,10#,12#,16#,20#,22#can reduce LDH leakage to varyingdegrees(P<0.05).Conclusion:The components of thermal transformation products from swertiamarin showthe hepatoprotective effect active and improve the cell survival rate of5mmol/L APinduced L-02at48h, and reduce the LDH leakage.
