Objective: Established the split method of nature and flavor material foundation ofCortex Mori. Chemical Characteristics of Cortex Mori split components and verifythat each component of taste and do not split the cross-cutting nature, to tastetraditional Chinese medicine can be split and combined of research ideas and herbaltaste objectivity of the scientific connotation of new hypotheses provide a scientificbasis.Methods:(1)Determine the content of total flavonoids by UV and determine thecontent of index constituent of Kuwanone G by HPLC. The optimum extractingcondition was investigated by orthogonal design and regard the quality of extractionas the investigated index.(2)This test using HPLC method established the specificChromatogram of Cortex Mori H2O-extract,and Kuwanone G,Morusin,mulberrosideA,Morin hydrate,Sanggenon D peak can be identificated. The column wasPLATISIL(TM))ODS C18(250mm×4.6mm,5μm). The mobile phase wasMethylalcohol-0.1%phosphoric acid. Nucleosides Min:0-60-90-100,CH3OH%:0-56-100-100;The column temperature was30℃. The flow rate was1.0mL/min. The detecting wavelength was set at280nm.(3)According to the chemicalcomposition of Cortex Mori,by steam distillation, extraction, alcohol, macroporousadsorption resin, such as technology, established the split method of nature and flavormaterial foundation of Cortex Mori. Using GC-MS, HPLC chromatography andstoichiometry to the nature and flavor detached composition of mutual validationstudies.Results:(1)The optimum extracting condition was A3B3C2with the quality of extraction of total flavonoids as the investigated index. The optimum extractingcondition was A2-3B1-3C1-3with the quality of extraction of Kuwanone G as theinvestigated index. With the requirement of industrial production, the optimumextracting condition was A3B3C2. That is adding ten times amount of water andrefluxing three times,1h every time.(2)established the specific Chromatogram ofCortex Mori H2O-extract,and Kuwanone G,Morusin,mulberroside A,Morinhydrate,Sanggenon D peak had been identificated. Through precision,stability,reproducibility and other methods, the relative retention time and relativepeak area RSD value is less than3%.(3)we splitted the material basis of decoction ofCortex Mori for7groups, which were30%Alcohol elution composition and50%Alcohol elution composition,80%Alcohol elution composition, alcohol-precipitationcomposition, alcohol-precipitation composition respectively. Of which30%,50%,80%ethanol elution compositions by HPLC-PDA, HPLC-ELSD, UPLC-DAD checkidentification and cluster analysis, principal component analysis and orthogonalpartial least squares analysis, finalized30%,50%,80%ethanol elution compositionssubstantially free of chemical components intersect.38components in Cortex Moriare detected by GC-MS and27compounds are identified by MS in combination withKovats retention index. The sum total of its relative content of compounds detected in93.79%. The compounds with high contents are as follows: Squalane (50.40%),α-Amyrin (10.86%), Methyl linoleate (9.33%), β-Amyrin (4.84%), Hexadecanoic acidmethyl ester (3.08%),11-Octadecynoic acid methyl ester (1.76%) and so on.Conclusion: The split technology of nature and flavor material foundation ofCortex Mori is stable and practical. Cortex Mori split component GC, HPLCcharacteristic patterns can reflect the characteristic taste of each chemicalcharacteristics of the components of the split. Indicated that Cortex Mori resolvabilityconstituent holo-composition which was detached uncross as each one could be wellseparated under no supervision mode. Offered the scientific base to the newhypothesis for the scientific connotation of the medicinal properties of the objectivityof provides.
