Purpose:Observation xylanase protein2(Aggrecanase2/ADAMTS-5) and tissue inhibitor of metalloproteinase3(tissue inhibitors of metalloproteinases3, TIMP-3) in diffirent human lumbar intervertebral disc degeneration,and discuss the role of them in nucleus back role in nucleus pulposus. Method:Using Pfirrmann classification method to class Intervertebral disc degeneration MRI.Divided into three groups: control group (Pfirrmann grade I-Ⅱ),degeneration group (Pfirrmann grade Ⅲ-IV), and severe degeneration group (Pfirrmann grade V). Collected45cases who acceptedremoval of the nucleus pulposus specimens from May,2012to December,2012in Guilin Medical lumbar spine surgery surgical in. Each group containn15cases. After formalin-fixed paraffin-embedded, then using immunohistochemistry to detect the nucleus pulpos us cells Aggrecanase2and TIMP-3expression. Result:The positive cell rates of Aggrecanase2in the Nucleus pulposus cell are13.58±7.76%,33.48±13.95and56.00±18.39%in the control group,degeneration group and severe degeneration group, respectively.Compared with the control group, expression of degeneration group is enhanced and the difference has statistics significance (P<0.001);compared with and the control group and degeneration group,expression of severe degeneration group is enhanced and the difference has statistics significance (P<0.001).The positive cellrates of TIMP-3are34.78±13.80%,46.77±10.98%and50.65±16.45%in the control group,degeneration group and severe degeneration group,respectively.Compared with the control group,the expression of degeneration group is enhanced and the difference has statistics significance (P<0.05);compared with the control group,the expression of severe degeneration group is enhanced and the difference has statistics significance (P<0.01);compared with degeneration group,the expression of severe degeneration group is enhanced and the difference has statistics significance (P<0.05).Aggrecanase2/TIMP-3ratio is45.37±30.57%,75.90±40.30%,120.62±49.67%in control group,degeneration group and severe degeneration group, respectively.Compared with the control group,the rat io of degeneration group is higher and the difference has statistics significance (P<0.05);compared with control group,the ratioof severe degeneration group is higher and the difference has statistics significance (P<0.001);compared with degeneration group,the ratio of severe degeneration group is higher and the difference has statistics significance (P<0.01).Conclusion:As the degree of the degeneration of nucleus pulposus increases,the expression of Aggrecanase2and Timp-3in the nucleus pulposus of Lumbar Intervertebral Disc and Aggrecanase2/timp-3ratio will rise,which indicates that these two are closely connected with the degeneration of nucleus pulposus, and there is positive correlation between its ratio and the degree of the degeneration of nucleus pulposus.
