Objective:To investigate the expression level of miRNA-181a in PBMCs and level of serum IL-1α of chronic obstructive pulmonary disease patients, as well as the correlation between miRNA-181a and IL-1α.Method:Patients of the Third Hospital of Xiangya from June to September of2013were divided in two groups, AECOPD group including19patients from inpatients and stable COPD group including23patients from outpatients. Meanwhile the health volunteers from Health Management Centre of the Third Hospital of Xiangya were divided into heavy smoker group with19subjects and nonsmoking health control group with16subjects. PBMCs expression of miRNA-181a of all subjects were determined by RT-qPCR and the serum level of IL-1α of all subjects were measured by ELISA. COPD relevant questionnaire and lung function conducted with COPD patients as well as other clinical index were conducted with in all subjects. Relevant statistical methoud were used to analyze the experimental and clinical data.Result:PBMCs level of miRNA-181a were decreased in COPD pacients including stable COPD group and AECOPD group compared with health control group(p<0.05). Lung function of COPD pacients is negatively relevant to their CCQ and MRC as well as CAT questionnaire score.The serum IL-1α level of AECOPD group is higher than health control group, stable COPD group and heavy smoker group(p<0.01). PBMCs level of miRNA-181α is negatively relevant to serum IL-1α level in COPD patients. Heavy smoking has a down regulative effect on expression of miRNA-181a, which is negatively relevant to lung function.Conclusion:1. PBMCs expression of miRNA-181a were significantly decreased in COPD patients compared with health control group.2. The level of serum IL-1α of AECOPD group is higher than health control group.3. PBMCs expression of miRNA-181a is negatively relevant to level of serum IL1a in COPD patients, miRNA-181a might have an regulative effect on expression of serum IL1a, thus plays an important role in progression of AECOPD.9figures,9tables,30references...
