Abstract/Summary:
Objective:To investigate The effects of sperm acrosin activity,seminal plasma zinc content and seminal plasma PMN-elastase level on IVF-ET.Method:Semen samples were obtained from153males who accept IVF-ET in our reproductive center of second Xiangya hospital between January2012and December2012.Females are infertility only for tubal factors and male’s semen analysis are nomal.1.Test sperm acrosin activity,seminal zinc concentration and seminal plasma PMN-elastase concentration,then to analysis the correlativity between the activity of acrosomal enzyme, the concentration of seminal zinc, the concentration of seminal plasma PMN-elastase and fertilization rate of IVF-ET.2. We devided the samples into2groups depend on the pregnancy outcomes, then to analysis the difference of sperm acrosin activity,seminal zinc concentration and seminal plasma PMN-elastase concentration between the two groups.3. comparison the different concentration of seminal plasma PMN-elastase whether influence the fertilization rate, the activity of sperm acrosin and the concentration of seminal zinc.Results:1. A positive correlation exists between the sperm acrosin activity and the fertilization rate,(r=0.316,p<0.05).The sperm acrosin activity was (85.42±24.68) uIU/106sperm in pregnancy group,(81.99±15.02) uIU/106sperm in nonpreganancy group, pairwise comparison was not statistically significant (t=0.779,p<0.05)2.No correlation between the concentration of seminal plasma zinc and the fertilization rate,(r=0.078,p>0.05). The the concentration of seminal plasma zinc was (6.05±2.86)μmol/ejaculation in pregnancy group,(5.82±2.64)μmol/ejaculation in nonpreganancy group, pairwise comparison was not statistically significant (t=0.390,p<0.05)3.A negative correlation exists between the concentration of seminal plasma PMN-elastase and the fertilization rate (r=-0.167,p <0.05). The seminal plasma PMN-elastase concentration was (745.57±93.87) ng/ml in pregnancy group,(775.411±107.68) ng/ml in nonpregnancy group, pairwise comparison was not statistically significant (u=1.428,p<0.05)4. According to the level of seminal plasma PMN-elastase,we divided the patients into uninfected group (the concentration of seminal plasma PMN-elastase<290ng/ml), recessive infection group (the concentration of seminal plasma PMN-elastase was290-1000ng/ml), confirmed infection group (the concentration of seminal plasma PMN-elastae>1000ng/ml).the three groups of fertilization rate was (84.02±24.99)%、(80.98±26.96)%and(74.01±23.12)%respectively.The difference of fertilization rate between the groups was statistically (F=3.78, p<0.05). The three groups of the sperm acrosin activity was (84.24±14.85)μIU/106sperm,(79.06±16.70)μIU/106sperm and (74.38±15.32)μIU/106sperm respectively. The difference of sperm acrosin activity between the groups was statistically (F=3.12,p<0.05). The three groups of the seminal plasma zinc concentration was (6.34±2.02) umol/ejaculation,(6.29±2.29) umol/ejaculation and (5.01±1.85) umol/ejaculation respectively, There was no statistically significant difference between the groups (F=2.141,p>0.05)Conclusion:1. The sperm acrosin activity reduced can lead to IVF fertility rate decreased, but has no effect on the pregnancy rate, sperm acrosin activity can be used as one of the indicators of selecting the fertilization method of vitro fertilization.2. The seminal zinc concentration has no effect on IVF fertilization rate and pregnancy rate.3. The increase of seminal plasma PMN-elastase concentration can decrease acrosin activity and IVF fertilization rate,but has no effect on the pregnancy rate.
