Objective:To compare the effect of proliferation, apoptosis and cell cycle on human osteosarcoma cell MG-63and U20-2by metformin and adriamycin.Methods:The cell viability was tested by MTT colorimetric assay, after MET and ADM were added together or alone, apoptosis percentage (AP) and cell cycle was assessed by flow cytometry (FCM).Results:MTT colorimetric analysis demonstrated that the inhibition effect of MET and ADM singly and both on the proliferation of MG-63and U-2OS cell was found in dose-and-time dependent manner(P<0.05). Flow cytometry showed that the MG-63cell cycle was induced by metformin at48hours, the ratio of G0/G1phase was increased and the ratio of S phase was decereased significantly(P<0.05). Meanwhile, both of the two drugs induced MG-63cell apoptosis, and combined use resulted in synergic effect.Conclusions:MET and ADM can depressed the proliferation of MG-63and U-2OS cell by dose-and-time-dependent. MET could also induce MG-63cell apotosis and arrest cell cycle at the phase of G0/G1. When MET is combined with ADM, the effect of proliferation and apoptosis is significantly enhanced.
