Objectives:Dectecting the expression and the methylation status of the SPINT2gene in the human gliomas of the different WHO classification, to explore the relationship between the methylation status of the SPINT2gene and the occurrence and development of the gliomas.Methods:Selecting24glioma specimens and8normal brain specimens, using the western blot method to detect the expression of the SPINT2gene; While using the methylation PCR (MSP) to detect the promoter methylation of the SPINT2gene in normal brain tissues and glioma tissues.Results:1. Using the western blot method to detect the expression of the SPINT2gene:The SPINT2protein between the normal brain tissues and the low-grade glioma tissues have statistical differences(P=0.008); The SPINT2protein between the normal brain tissues and the high-grade glioma tissues have statistical differences(P=0.000); The SPINT2protein between the low-grade glioma tissues and the high-grade glioma tissues do not have statistical differences(P=0.061).2. Using the Methylation Specific PCR method to detect the Methylation status of the SPINT2gene:The rate of the methylation of the SPINT2gene between the normal brain tissues and the low-grade glioma tissues do not have statistical differences(P=0.347); The rate of the methylation of the SPINT2gene between the normal brain tissues and the high-grade glioma tissues have statistical differences(P=0.024); The rate of the methylation of the SPINT2gene between the low-grade glioma tissues and the high-grade glioma tissues do not have statistical differences(P=0.097).Conclusions:1. The SPINT2expressed in both the normal brain tissues and the different levels of glioma tissues. The SPINT2protein of the normal brain tissues is more than all pathological classifications of gliomas; but there is not obvious difference between the low-grade glioma tissues and the high-grade glioma tissues.2. Methylation of the SPINT2gene can not be found in the normal brain tissues. The methylation rate of the normal brain tissues is less than the high-grade glioma tissues. But there is not obvious difference between the normal brain tissues and the low-grade glioma tissues, the low-grade glioma tissues and the high-grade glioma tissues.
