Purpose:By measuring the content of IL-10、IL-1β、ICAM-1and NF-κB to observe themechanism of H2S play a protection on lung through adjust inflammatory cytokines and hoping toprovide the theoretical basis for clinical treatment of paraquat poisoning.Methods:Sixty healthy rats were randomly divided into6groups.The blank group:NaCl group(intraperitoneal injection of0.9%NaCl)、The NaHS group (intraperitoneal injection ofNaHS);Experimental groups:PQ group、PQ+low dose of NaHS(L group0.78mg/kg)、PQ+middledose of NaHS(M group1.56mg/kg)、PQ+high dose of NaHS(H group3.12mg/kg).All rats werebleeded to death after PQ lavaged24hours;Observing the lung wet/dry weight,right lung tissuestained with HE,at high magnification observed the changes of pathological score of lung injury.Three experimental analysis methods (Western Blot、ELISA and RT-PCR) were respectively usedto measure the expression of NF-κB protein in the lung tissue、the content of IL-1βand IL-10inthe plasma of rats、the level of IL-1β、IL-10and ICAM-1mRNA in the lung tissueResults:1. General activity in rats、lung tissue pathology change and (IQA) score:At the optical microscope observed the HE staining of lung tissue section,between NaClgroup and NaHS group did not see the pathological change,IQA had not obvious difference(P>0.05).The lung tissue structure of PQ group was obviously damaged,there were a large number ofalveolar interval fracture and mutual fusion, many red blood cells and inflammatory cells in thealveolar,meanwhile alveolar septum visible inflammatory cell infiltration and septa in lung tissuechanged into thickening,some of alveolar transparent membrane formation,IQA and(W/D)in PQgroup was increased had statistically significantly (P<0.01).In L、M and H groups most ofstructure of alveolar were intact and IQA、(W/D)was statistically significantly lower than PQgroup (P<0.01) and the level of decrease was positively correlated with the concentration ofH2S,but compare with the NaCl and NaHS groups,There were still obviously damaged. 2.The content of IL-10、IL-1β、ICAM-1、NF-κBCompared with NaCl group rats,NaHS group rats the content of IL-1β、IL-10in the plasmaand IL-1βmRNA、IL-10mRNA、ICAM-1mRNA、NF-κB in the lung tissue had not obviousdifference(P>0.05).PQ goup rats the content of IL-1βin the plasma and IL-1βmRNA、ICAM-1mRNA、 NF-κ B in the lung tissue were higher than NaCl group rats (P<0.01).Compared with PQ group rats,the L、M、H group rats the content of IL-1βin the plasmaand IL-1βmRNA、ICAM-1mRNA、NF-κB in the lung tissue were lower than PQ group rats,the content of IL-10and IL-10mRNA are higher than PQ group rats (P<0.01)Conclusions:1.Increasing exogenous H2S can decrease rats acute lung injury pathological scores,bothpulmonary edema and inflammation were decreased in early. The level of decrease was positivelycorrelated with the concentration of H2S2.H2S can suppress the expression of ICAM-1mRNA、IL-1β mRNA and up-regulate the expressionof IL-10mRNA,further more it can inhibit the level of NF-κB protein expression,through thismechanism to protect lung tissue...
